I'm happy to share the newest exploits of my lab with you all! Spearheaded by Dr. Bhaskar Chatterjee, this study focuses on the development of small molecular inhibitors of the human AMPylase, FICD. We disclose and discuss a first set of FICD inhibitors with proven activity in intact cells. We use molecular modeling to define the binding mode, determine the molecule’s impact on both FICD-mediated protein AMPylation and deAMPylation in distinct cell types, and show that the molecules are active against wild-type and disease-associated mutants of FICD. We also demonstrate that the lead molecule is amendable to medicinal chemistry and describe the stunningly beneficial effect of the molecules on pro-insulin processing in beta cells.
