We are thrilled to #announce the publication of our latest research article titled "The potential role and mechanism of circRNAs in Ferroptosis: a comprehensive review" in the esteemed journal,Pathology Researchand Practice, Impact Factor: 2.8. This #comprehensive_review delves into the intriguing world of #cell #death #mechanisms, with a particular focus on Ferroptosis - a unique, #iron_dependent form of regulated #cell_death distinguished by its reliance on reactive oxygen species (#ROS) and distinct from other forms such as #necrosis and #apoptosis. Our study highlights the crucial regulatory roles of circular #RNAs #(circRNAs) within ferroptosis-mediated pathways, underscoring their significance in #diseases linked to ferroptosis including #cancer, #neurological, and #cardiovascular diseases. Through our research, we offer new insights into the #diagnosis, #treatment, and #prognosis of ferroptosis-related diseases, emphasizing the therapeutic implications of targeting circRNAs in cancer treatment and the modulation of ferroptosis #pathways. This opens up novel avenues for advancements in disease management and highlights the potential of circRNAs as both #diagnostic markers and #therapeutic targets. Athira Pharma has announced the results of its Phase II/III LIFT-AD trial evaluating fosgonimeton, an HGF/MET pathway modulator, for the treatment of mild-to-moderate Alzheimer's disease. The trial did not meet its primary or key secondary endpoints. Key Findings: Primary Endpoint: The Global Statistical Test (GST) showed a non-significant change of -0.08 for fosgonimeton compared to placebo. Cognition: The change in ADAS-Cog11 scores was -1.09 for fosgonimeton versus -0.39 for placebo. Daily Functioning: The ADCS-ADL23 score increased by 0.65 with fosgonimeton, while the placebo group saw a decline of -0.02. While the primary endpoints were not achieved, the trial's biomarker data showed potential positive trends, including reductions in plasma p-Tau217 levels, which are indicative of Alzheimer's disease pathology. Subgroup analyses also suggested that fosgonimeton may offer benefits to specific patient populations, such as those with moderate Alzheimer's or APOE4 carriers. #ClinicalTrials #AlzheimersDisease #PharmaceuticalResearch #Neuroscience #DrugDiscovery

Vaxxinity, unveiled intriguing findings at the 2024 American Academy of Neurology conference concerning their investigational active immunotherapy, UB-312. The study aimed to assess target engagement in cerebrospinal fluid (CSF) samples of Parkinson's disease (PD) patients using the Amprion R&D aSyn seed amplification assay (aSyn-SAA). UB-312 represents a promising direction in PD treatment by targeting pathological forms of alpha-synuclein (aSyn), a pivotal protein linked to the advancement of synucleinopathies. Unlike traditional methods, UB-312 endeavors to stimulate antibodies that specifically identify and counteract these detrimental aSyn variants, potentially arresting or decelerating the neurodegenerative mechanisms underlying PD. A paper, published in Nature Immunology, discusses TREM1, an immunoreceptor that belongs to the immunoglobulin superfamily, which is expressed on myeloid cells, including peripheral macrophages and microglia. Known to amplify myeloid-mediated inflammatory responses triggered by pathogen challenges, such as bacterial and fungal infections, through the release of pro-inflammatory chemokines and cytokines, TREM1+ microglia have now been found to assemble at sites of β-amyloid deposition in the brains of patients with Alzheimer's disease (AD) and related mouse models of cerebral amyloid accumulation and contribute to neuroinflammation in AD. A Biological Definition of Parkinson’s Disease: Two Proposals Pave the Way Just as fluid biomarkers for amyloidosis and tau have revolutionized clinical studies of Alzheimer’s disease, α-synuclein markers are poised to do the same for Parkinson’s. With the advent of sensitive assays that reflect α-synuclein pathology brewing in the brain, the time is ripe to center our view of PD and related disorders around their underlying biology, argue a Personal View and a separate Position Paper in the February issue of Lancet Neurology. Each proposes a system to classify and, in one case, also stage Parkinson’s and related diseases based on their pathology. Both hope that shifting toward a biology-based framework will help scientists zero in on specific mechanisms underlying disease, better-characterize people with parkinsonism, and streamline therapeutic development.
