Our latest pre-print in BioRxiv. Here, we analyze the effects of the pharmacological or genetic disruption of ER aminopeptidase 1 (ERAP1) on the immunopeptidome, peptidome and metabolic state of cancer cells. ERAP1 is an important enzyme that regulates adaptive immunity and is currently a pharmacological target for cancer immunotherapy. While ERAP1 is mostly studied for its ability to regulate the cellular immunopeptidome, other functions have also been proposed. Here, by using a combination of proteomic and biochemical analyses, we show that modulation of ERAP1 activity can generate unique immunopeptidomes, mainly due to altered peptide processing in the ER, but also induce changes in the cellular proteome and metabolic state which may have further effects on tumor cells.
