Have you seen the latest publication in Cells MDPI by our esteemed #ISSLS member Stefan Dudli and his co-authors? 🔥 This novel study aimed to identify #TLRs on CEPCs and their role in pro-inflammatory and catabolic gene expression, revealing some important findings that could impact the future of spinal health. https://lnkd.in/gUHB_Vau This research underscores the significance of TLRs, particularly TLR2, in the CEP, as its higher cell density could cause strong local pro-inflammatory changes. In a field where the mechanisms linking CEP degeneration to DD and MC remain unclear, these advancements are crucial for understanding how biological degenerative changes can affect the function and stability of CEP contributing to DD, MC, and ultimately LBP. Key findings: >  This study demonstrated that the cartilage endplate has critical pro-inflammatory and degenerative functions and hence is not just a biomechanically and biophysically important structure, but also biologically. >  Considering the about 4-7x higher cell density in the cartilage endplate than the intervertebral disc, understanding the biological role of the endplate in disc degeneration and Modic changes is highly important.  >  The authors showed enhanced TLR2 expression in cartilage endplates. This had pro-inflammatory and pro-catabolic consequences leading to endplate resorption, a critical factor in disc degeneration and Modic changes. This publication is a testament to the incredible work being done by #ISSLS members. In this paper we used unsupervised learning and archetype analysis to explore the alternative splicing landscape of high-risk Wilms’ tumors. We identified a set of transcripts which are alternatively spliced between different regions of gene expression latent space. Many of these transcripts were found to be associated with the epithelial to mesenchymal transition (EMT) and muscle development. Our findings provide new insights into the etiology of Wilms’ tumors and suggest that specific splicing mechanisms in early stages of development may contribute to tumor development in different patients.  See more at https://lnkd.in/dpGq-piX

The future of back pain treatment is here - Nanocarriers Deliver Gene Therapy Scientists have made a big step forward in treating chronic back pain. They used tiny, engineered particles called extracellular vesicles (EVs) carrying the gene FOXF1 to repair damaged spinal discs in mice. This new, minimally invasive approach could not only help with back pain but also other joint problems. More research is needed, but the results so far are very promising. Hypertrophic cardiomyopathy (HCM) affects between 600,000 and 1.5 million Americans, but the complexity of the condition has made finding effective treatments challenging. The tide could change soon, thanks to research conducted at the Allen Institute for Cell Science. Researchers there have developed six pluripotent stem cell lines, each of which has a different key gene mutation found in HCM. These findings are helping scientists make generalizations about the disease, which can be used to develop more personalized treatment approaches. 📢 New pre-print out! I'm delighted to have played a part in this collaborative effort with the group of Carlos Parras at the Institut du Cerveau – Paris Brain Institute. With Jean-Baptiste Huré, Ph.D, we were able to implement my previous ex-vivo model to study the pro-oligodendrogenic properties of small bioactive molecules. Through a combination of in vitro, ex vivo and in vivo approaches, we demonstrated the ability of leucovorin and dyclonine to promote oligodendrogenesis and myelin repair, paving the way for a potential therapy for #MultipleSclerosis .
