In sleep-related clinical drug development, understanding the relationship between cognition, sedation, and wakefulness is pivotal for safety and efficacy decision making. Gather insights from Cogstate’s CIO, Paul Maruff, describing how cognitive assessments are used to inform decisions about mechanism of action, dosing, length of effect, and consequences on thinking of new therapeutic compounds. EEG can also be a valuable support to sleep exploration. Thanks to a reliable assessment of sleep latency, sleep EEG data can be used to characterise drug efficacy at an early stage. Don't miss the Alkermes presentation, which is a perfect example of this. #SLEEP2024
Today we announced new clinical data from the full #narcolepsy type 1 cohort of our phase 1b study evaluating our investigational orexin 2 receptor agonist in patients with #narcolepsy type 1. We look forward to presenting this data at #SLEEP2024, a joint meeting of American Academy of Sleep Medicine and Sleep Research Society. Read the press release: https://lnkd.in/gKd29Twe

Suggested read here for sleep medicine clinicians ⬇️ The PAP-RES algorithm is a new approach to CPAP therapy. The algorithm focuses on identifying OSA phenotypes, which are distinct groups of individuals with OSA who have different symptom burdens and cardiovascular vulnerability levels. This approach aims to personalize CPAP therapy based on the individual's phenotype, leading to better outcomes and a more effective treatment plan. Sham-controlled trials are needed to characterize the effect of hypoglossal nerve stimulation (HGNS) therapy on cardiovascular end points in patients with moderate-severe obstructive sleep apnea (OSA). What is the effect of HGNS therapy on measures of cardiovascular health in patients with moderate-severe OSA? In this double-blind, sham-controlled, randomized crossover therapy trial of 60 patients, mean 24-hour systolic blood pressure and other cardiovascular end points were not significantly different between sham and active HGNS therapy. Long term effects of #CPAP therapy for patients with obstructive #sleepapnea are still an open question Read my commentary in the Journal of Clinical Sleep Medicine on the latest review article on this topic. Results from the review article point to the lack of high-quality evidence from randomized controlled trials supporting the effectiveness of CPAP in reducing long-term adverse health outcomes, such as mortality, major cardiovascular events like stroke and myocardial infarction, and the development of conditions like anxiety. What are your thoughts on these surprising findings? My commentary: https://lnkd.in/d_X_SucU Review article: https://lnkd.in/dnmNF-GJ #sleep #sleephealth #sleepmedicine #education #science #research

Is transcutaneous electrical nerve stimulation (TENS) effective in reducing acute and chronic pain? Neuromodulation techniques such as TENS and BMT are used as a complementary therapy to alleviate sensations of pain, muscle tension and spasm, and the impact of an 'overprotective brain'. Patients in this study found that TENS provided benefits including enhanced function, improved psychological wellbeing, better sleep, and lower pain intensity immediately after TENS administering. As TENS treatments can be used in a clinical or at-home setting, patients found repeat treatments aided toward a quicker return to activities of daily living and improvements in quality of life. Melatonin is a powerful molecule, with actions throughout the body. Although it often comes to mind for sleep, it has more actions beyond sleep: for circadian health and chronic disease. Melatonin is strongly suppressed by light, and the darkness deficiency of the modern built environment. Happy to team up with colleagues to present the "Light and Melatonin as Medicine" masterclass through HealthMasters Live. Learn how to use these therapies from a range of experts in this 17-hour on-line class. You'll come away with many clinical strategies, and be able to use melatonin with precision. #melatonin #sleep #circadianrhythm #artificiallightatnight #CME https://lnkd.in/gC5YtQGY
