Excited to share that a paper stemming from my PhD project, titled "BDNF sensitizes bone and joint afferent neurons at different stages of MIA-induced osteoarthritis", has been published in Bone Journal. This work was supported by funding from the Australian National Health and Medical Research Council (NHMRC) and the University of Melbourne School of Biomedical Sciences Early and Mid-Career Association Collaborative Research Award. Our research sheds light on the critical role of BDNF/TrkB signaling in osteoarthritis (OA) pain. By demonstrating that TrkB is expressed in neurons innervating the knee joint and bone, and that BDNF application enhances their mechanical sensitivity, we provide valuable insights into the mechanisms underlying OA pain. Additionally, our findings reveal how BDNF expression changes throughout the disease progression, suggesting a potential therapeutic window for intervention. Importantly, TrkB inhibition effectively reverses sensitivity alterations at different stages of OA, indicating that targeting this pathway could offer new strategies for managing OA pain and improving patients' lives. Brown tumors (BTs) are benign, lytic lesions that result from long-standing untreated hyperparathyroidism. BTs as the initial manifestation of primary hyperparathyroidism are rare, especially in developed countries. The main differential diagnosis is of a giant cell lesion, which shares the same clinical, radiological, and histological features as BT. It is important to distinguish between these two bone pathologies in view of different treatment strategies; BTs often respond to correction of long-standing hypercalcemia whereas giant cell lesions often require surgical removal.
