Very comprehensive and insightful summary for the T-cell dysfunctions. We may need to consider that post translational modifications (PTMs) are involved in all the cases of T-cell dysfunction, in this case, methylation. PTM may have signaling roles of initiation and termination to functional proteins, eventually to cells. It is interesting that Th17 cells succeed baton from Treg in case of instability via RORrt. Indeed the roles of FOXO3 are also interesting because they will have lots of potentials of transcriptional activities. In this article, we may need to consider three chimeric factors (PTM, FOXO3 and RORrt) for maximizing T-cell therapies.

Inflammation models, reflecting crucial pathological aspects of human diseases, serve as a comprehensive platform for advancing preclinical drug development. The ability to assess therapeutics from ex vivo models through to more intricate in vivo models represents an essential tool in modern drug discovery.
