About a year ago a bleak review paper (Adkar-p et. al, 2023) alerted cultivators of a great threat to the Cannabis Industry - Hop Latent Viroid (HLVd), breaching the relatively small scientist audience and reaching mjbizdaily and the Wall Street Journal. Viroids are (or were thought to be; Lee et. al 2023) exclusive pathogens of plants, composed solely of small, covalently closed circular (ccc) RNA molecules of 220 to 450 nucleotides, with no protective lipid/protein cover as seen in viruses. Such simple elements are expected to lack specificity (as there are no determinants to restrict infections in specific plant species) and other biomolecular mechanisms to facilitate cell entry (like the Spike (S) glycoprotein of SARS-CoV-2). However, lacking such elements also renders the pathogens very resistant to plant defense responses. Since no exogenous molecules such as viral capsid proteins/viral enzymes are required for viroids, there is no efficient pathway to stop its proliferation by targeting these elements and prevent its passive/carrier facilitated passage through plasmodesmata and into new cells. Viroids hijack cellular enzymes for replication in the nucleus or chloroplast, depending on the viroid species, infecting entire plants in several days following introduction, which can occur mechanically by contaminated tools, insects, pollen, seeds, parasitic plants, shoot/root grafting, and fungal phytopathogens. As such, viroids are persistent and very successful plant pathogens, infecting between 90-100% of the hop samples tested throughout Europe (Faggioli et. al 2017). Hops may be infected for a long duration without any apparent symptoms, some plants will eventually (hence “latent”) show signs of “dudding disease”, which may cause yellowing, smaller inflorescence, and reduced glandular trichome densities and lower terpene concentrations in Hops, although this was found in a single study, and significant effects were observed in a single cultivar (Barbara et al,1990). In cannabis, two papers (Bektas et. al 2019, Warren et. al 2019) published back-to-back in the same journal used similar molecular methods to assess symptomatic and asymptomatic plants exhibiting brittle stems, an outwardly horizontal plant structure, stunting, malformation or chlorosis of leaves, and reduced flower mass and trichomes. Unlike hops, which may harbor the viroid while remaining asymptomatic, both articles found a high correlation between symptoms and HLVd-positive plants, with asymptomatic plants testing negative for the viroid in both studies. These findings were reported by a couple of online articles in January and September 2021 (HempIndustryDailyTM; Cannabis Business Times), also cited in last year’s review. taking into account the reported impact on cannabis yielded a profound value of ~5 billion dollars (potential) annual losses due to HLVd. Despite the grave potential threat to the industry, only a handful of scientific publications have cited last year’s review article by Adkar-Purushothama et. al. Of those, most were review articles describing various microbial and viral pathogens of cannabis (MIotti et al, 2023; Payment & Cvetkovska, 2023; Punja et al. 2023; Buirs & Punja, 2024). Only two articles have been published so far which provide experimental results of HLVd-cannabis interactions. In June of 2023, A paper describing a new viroid detection method dubbed MFDetectTM was published by Marti et. al involving authors from UC Berkeley, MyFloraDNA, Inc, and Glass House Farms. The method relies on Reverse Transcription of RNA using specific primers, followed by quantification utilizing dye-probes. The authors claim the method is significantly more cost-effective than the routinely used RT-qPCR, and is apparently on route to commercialization, as several authors declare conflicting interests. In December 2023, Attalla et al. reported findings on HLVd infection dynamics and transfer routes in hemp and other plants. The researchers assessed the genetic diversity of HLVd from 13 samples of infected hemp plants, finding substantial sequence diversity between isolates, with some conserved nucleotides at certain positions which allowed mapping the phylogenetic relationship between them. Additionally, the authors showed equal infection efficiencies of crude sap or RNA extract from infected plants, and pathogen distribution dynamics in plants up to 14 days post infection, showing the time-depended dispersion of the viroid within the plants. At 3-5 days post infection, less than 10% of the top leaves tested positive for HLVd, rising to ~80% after 7 days and 100% after 14 days. Additionally, ~70% of the root tissue tested positive after two weeks. The study also determined HLVd is highly transmissible through hemp seeds, at rates of 58 to 80%. Host range assays showed that tomato, cucumber, chrysanthemum, N. benthamiana, and A. thaliana are susceptible to HLVd infections, although all but A. thaliana exhibited no symptoms, and showed variable plant-part susceptibilities. Viroids are hypothesized to impact plants through RNA silencing when specific viroid sequence corresponds to host mRNA. When viroids are degraded, viroid fragments can interact with cellular enzymes which function to degrade similar sequences, thereby interfering with key cellular functions. In asymptomatic infections, it is possible that viroid RNA does not correspond to critical host mRNA sequences, or, that it is not degraded at sufficient rates and does not represent a significant energetic toll on the plant. The high genetic variability of HLVd found in the study by Atallah et al may suggest that viroids may turn virulent under some conditions (mutations), or for specific cultivars which are more reliant on proteins which can be silenced by degraded viroids.
