As highlighted by QbD principles, scale down system are commonly used in pharmaceutical process development to study the effect of manufacturing Critical Process Parameters (CPPs) on drug product Critical Quality Attributes in laboratory scale. However, translating manufacturing CPPs in small scale is often a challenge and might lead to undesired results if done improperly. This study investigates how stopper positioning and geometry influence mass transport resistance during the freeze-drying process, using Computational Fluid Dynamics (CFD). We analyzed conventional batch freeze-drying, continuous freeze-drying, and low-fill-volume drug products, highlighting the significant impact of stopper configurations on process variables, particularly for low-fill-volume formulations. Study of Hydrodynamic Stress in Cell Culture Bioreactors via Lattice-Boltzmann CFD Simulations Supported by Micro-Probe Shear Stress Method Mammalian cell cultivation in pharmaceutical industry can last up to units of weeks and requires proper transport of nutrients and oxygen for cell growth and production. Given the long time period, cells experience flow fields from all bioreactor’s zones, where the energy dissipation rate (ε) varies substantially. For the very first time, the micro-probe method is applied successfully not only to laboratory (3 L) and pilot scale (80 L and 200 L), but also to industrial production scale bioreactor (12,500 L) with Rushton turbine and pitched-blade (RT-PB) impeller configuration. Experimentally obtained data are used for the validation of comprehensive CFD scale-up study, using the Lattice-Boltzmann large eddy simulation (LB-LES) method. https://lnkd.in/gQy-_CbE #aspenalert #biotech #bioprocess

🔬 New Research Alert! published in #DrugDevIndPharm. 🌟 " Design, optimization and pharmaceutical characterization of wound healing film dressings with chloramphenicol and ibuprofen " 🌟 This research article focuses on the optimization of a film formulation for wound healing dressings. The study uses the Design of Experiments (DOE) approach to investigate the impact of various factors on the mechanical properties of the films. The aim is to predict an optimal formulation based on these results. The article also discusses the testing of the final product in terms of film dimensions, adhesive properties, mechanical profile, and other characteristics such as tensile properties, swelling behavior, Water vapor transmission rate (WVtR), surface morphology, thermal behavior, wettability, in vitro release of APIs, in vitro cell viability, and antibacterial activity.
