Come get your assay on with us! If you’ve never designed one yourself or considered how dPCR assays are designed and developed, the latest Absolute Gene-ius episode will help you appreciate the skill and artistry involved. We’re joined by two PCR assay design pros - Kimi Soohoo-Ong, and Dr. Rounak Feigelman, both from Thermo Fisher Scientific – and they shine a light on the many factors that must be considered to design a winning PCR assay. From the level of fragmentation of nucleic acids in the sample, to what other species’ genomes that may be present in the sample, to what the sample matrix may contain, to the PCR master mix being used, if multiplexing is required, to what assay controls will be, and more! These two practiced bioinformaticians cover these challenges and then tell us how their team overcomes challenges to develop winning assays for both qPCR and dPCR applications. Learn with IDT! ✍ #Multiplexing, when done right, can leverage your platform to increase your capacity and your efficiency. In #multiplexqPCR, multiple targets are amplified in a single reaction tube. Each target is amplified by a different set of primers and a distinctly labeled probe that will distinguish each #PCRamplicon. Multiplexing provides some advantages over single-reaction #PCR, including requiring an overall lower amount of starting material, increased throughput, lowered reagent costs, and less sample handling. However, the experimental design for multiplexing is more complicated because the amplification of each target can affect others in the same reaction. Therefore, careful consideration of the design of the reaction is critical. Want to see what we suggest for a multiplex qPCR approach? Do you work on the RAS signalling pathway? We have an on-demand webinar which talked about how to quantify RAS drug-target engagement in live cells? During the webinar, Dr Matt Robers (Sr Research Scientist, Promega Corporation) discussed the impact of oncogenic KRAS on engagement potency of downstream effectors, including BRAF. Dr Robers demonstrated how various allosteric and orthosteric engagement mechanisms could be detected using pan-KRAS NanoBRET™ technology. Life of Molecular Diagnostic Tests Beyond RT-PCR: This Saturday’s Business IDEA The COVID-19 pandemic introduced the RT-PCR test to the masses, making it a mandatory requirement for travel within and between states, even for those without a scientific background. But is RT-PCR the final frontier, or can other tests, which are more affordable, be made functional and accessible in resource-limited areas? Specifically, can they be brought to every corner of the African continent? Yes, there are, though they may be few in number, they have the potential to bring about change. One such test is the Recombinase-Aided Amplification (RAA) test, which offers a promising alternative. #RecombinaseAidedAmplificationAssay #RTPCRvsRAA #MolecularDiagnostics #PathogenDetection #PointOfCareDiagnostics #NucleicAcidAmplification #Biotechnology #HealthcareTechnology
