Electrophysiology serves as a fundamental tool in the investigation of epilepsy, providing valuable insights into the underlying mechanisms of this complex neurological disorder. At EphyX Neuroscience, we routinely employ a range of epilepsy in vitro models including transgenic rodents and/or pharmacologically-induced epileptiform activity to test the effects of drug candidates. Below is a short example of what can be expected qualitatively from single cell recording of a CA1 pyramidal cell and pharmacological induction of epileptiform activity in P30 C57/Bl6 mouse acute brain slice using a combination of low magnesium and gabazine. The top panel (A) shows the spontaneous bursts of action potentials (APs) elicited in this condition compared to baseline standard ACSF. The bottom panel (B) shows the hyperexcitability of the cell when eliciting action potentials. Typical analysis would focus on the APs frequency, threshold, height and half-width but we adapt to your needs to extract and analyse the most relevant parameters for your project. 🧠 Epilepsy Insights with Brain Slice Electrophysiology 🧠 At EphyX Neuroscience, we leverage brain slice electrophysiology to create a physiologically relevant environment for studying various pathological conditions and evaluating the impact of your lead compounds on neuron-specific activities. This approach yields highly valuable functional insights. For example, we routinely employ a range of in vitro models to assess the potential of test compounds and their mechanisms of action in the context of epileptic seizures. One notable model utilizes Oxotremorine, a muscarinic acetylcholine receptor agonist that mimics the hyperactivity observed during epileptic seizures. By selectively activating cholinergic pathways, Oxotremorine enhances neuronal intrinsic excitability. This effect is clearly demonstrated through whole-cell patch-clamp recordings of a CA1 pyramidal cell before and after Oxotremorine application: increased firing frequency for a given current injection step, and the generation of spontaneous bursts of action potentials.
