This time we are happy to announce a scientific publication in #MolecularPharmaceutics, a journal of the American Chemical Society, on another tracer that has recently become part of our portfolio: TRAP-WL12 for imaging of #PDL1 expression.   The team (Neil Quigley, Katja Steiger, Stefanie Färber, Frauke Richter and Wilko Weichert) led by TRIMT’s CSO Dr. Johannes Notni investigated the PD-L1 targeting PET tracer Ga-68-TRAP-#WL12 in murine xenografts of nontransfected PD-L1 expressing tumor cell lines. They showed that tumor uptake of the tracer was high demonstrated by the detection of even very low PD-L1 expression densities with high sensitivity and specificity, accompanied by a favorable biodistribution profile with rapid clearance from the blood pool, renal excretion, and low organ background. Today, we had the honor to present our Digital Pathology and AI solutions to enhance the understanding of fibrosis and inflammation in the context of Extracellular Matrix (ECM) Pharmacology at the 2024 ECM Congress in Copenhagen. Quoting Prof. Morten Asser Karsdal, Chair of the congress: "Dysregulation of the ECM, characterized by either elevated degradation or formation of tissues, is a universal characteristic observed in many chronic diseases. To make a real impact on the lives of patients, we need to learn from other diseases where fibrosis and tissue destruction are central mechanisms involved in the pathophysiology." Are you doing proteomics correctly for xenografts? Accurate biomarker analysis is crucial for identifying predictive biomarkers and making informed decisions before moving to clinical trials. However, PDX models, while predictive, pose challenges. Once human tumors are implanted, mouse stroma quickly replaces human stroma, mixing signals and complicating analysis. #CureDuchenne is pleased to share that Wave Life Sciences announced positive interim data from the ongoing Phase 2 FORWARD-53 study in Duchenne amenable skipping exon 53. After 24 weeks of 10 mg/kg dosing every two weeks, individuals treated with WVE-N531 demonstrated an average dystrophin expression of 9.0% of normal when adjusted for muscle content (5.5% of normal dystrophin if unadjusted). Muscle biopsies also showed signs of improvement in muscle health (myofiber size and diameter) and the presence of the drug in muscle satellite cells. Serum biomarkers, such as creatine kinase (CK), were also reduced. Wave plans to complete the 48-week FORWARD-53 trial in Q1 2025 and receive feedback from regulators about a pathway to accelerated approval. Excited to share our comprehensive review of the latest hybrid imaging techniques for the diagnosis, staging, and therapeutic management of neuroendocrine tumors (NETs). Key highlights: - 68Ga-DOTA-peptide PET/CT is the current gold standard, enabling lesion detection with high sensitivity (90-98%) and specificity (92-98%) - 68Ga-DOTA-peptide uptake can stratify prognosis and select patients for peptide receptor radionuclide therapy (PRRT) - Emerging PET/MRI offers simultaneous anatomic and functional imaging with improved lesion detection and characterization compared to PET/CT - Promising new molecules include 18F and 64Cu-labeled somatostatin receptor agonists and antagonists Further research should refine response assessment criteria and standardize reporting guidelines to maximize clinical impact. The bystander effect of ADCs needs to be considered when designing biomarkers. Trastuzumab deruxtecan (T-DXd) is a HER2-directed humanized #ADC that has a bystander effect on HER2-negative cells that are proximal to HER2-positive cells. By mapping the proximity of HER2-negative cells to HER2-positive cells, you can infer how the bystander antitumor effect may affect a tumor, creating a better predictive biomarker. This method initially requires estimating the distribution of #HER2 protein expressed by each breast tumor cell to consider cellular heterogeneity. It alone is a much better way to assess HER2 than the categorical estimates currently approved by the FDA which uses IHC scores of 1, 2, and 3. Much AI-based image analysis is trying to use the existing pathology models as predicate devices, to tread an easier path to approval.
