Due to their size and complexity, degraders utilized in targeted protein degradation (TPD) create a need for approaches that streamline the design and study of these heterobifunctional molecules.
To support research and development of TPD therapeutics, new databases containing fragments specific for designing linkers of degraders have been created in Spark™, and can be used for bioisostere replacement or ligand joining experiments.
We explore how these new linker databases comprehensively include known and commercial degrader linkers as supported by experimental data, and demonstrate how Spark is a promising tool for approaching linker design for heterobifunctional degraders.
