Source: Talanta This study presents a new methodology combining differential mobility spectrometry (DMS) with liquid chromatography-tandem mass spectrometry (LC-MS/MS) to overcome challenges in quantifying limaprost. The method achieves high sensitivity and selectivity, making it suitable for pharmacokinetic and bioequivalence investigations. The integration of DMS with LC-MS/MS is shown to be effective in analyzing biological samples with multiple interferences.

Source: Analytical chemistry This article presents a sensitive and comprehensive method for analyzing signaling lipids using targeted liquid chromatography-mass spectrometry (LC-MS/MS). The method overcomes challenges such as a wide dynamic range, isomeric and isobaric species, and unwanted interaction along the separation path. It allows for the monitoring of 388 lipids in a single run of only 20 minutes and has been successfully applied to analyze the human plasma signaling lipidome and investigate lipid signaling pathways during platelet activation. This method provides unprecedentedly comprehensive analysis and direct insight into signaling pathways.

We are excited to announce the recent publication of our customers' study in Journal of Chromatography A, titled "In-depth Characterization of a Cysteine-Linked ADC Disitamab Vedotin by Multiple LC-MS Analysis Methods and Cutting-Edge Imaged Capillary Isoelectric Focusing Coupled with Native Mass Spectrometry." This research presents a state-of-the-art iCIEF-MS technique under native conditions for detailed analysis of the cysteine-linked ADC, Disitamab Vedotin, in conjunction with traditional online LC-MS approaches. The iCIEF-native MS method preserves the native state of the ADC during both iCIEF separation and MS detection, while also enabling straightforward method transfer from development to quality control (QC). In a recent publication, Merck researchers discovered the importance of incorporating flow into preclinical in vitro models. Quantification of virus like particles (VLPs) is challenging due to the large size and structural heterogeneity of the molecule. Structural heterogeneity arises due to random assemblage of L1 protein with other host cell proteins. In this study, we propose use of Reversed phase chromatography for separation and identification of VLP L1 proteins in inhouse crude and purified samples and quantification of L1 protein at different process points. We are delighted to announce that on April 5, 2024, the results of an observational study comparing a brand-new point-of-care fluorescence immunoassay for determining circulating levels of vitamin D with ultra-high performance liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS) were published in the prestigious peer-reviewed journal "Molecules." Vitamin D deficiency and insufficiency, which can lead to many pathological conditions, have become a widespread health problem, resulting in an increase in requests for determining circulating levels of 25-hydroxyvitamin D (25-OH-D), that is currently the main indicator of vitamin D status and it is performed by different assays. However, the analytical performance of these immunoassays is highly variable and does not necessarily provide comparable results. To address this issue, considerable efforts have been made to improve the metrological performance of this test to achieve comparable results across different methods and manufacturers. In particular, the AFIAS Vit D assay, a point-of-care fluorescence immunoassay based on a single cartridge with a lateral flow architecture and pre-filled with all the necessary reagents for the test, showed a good correlation with LC-MS/MS when running on both the Afias-1 and Afias-10 analyzers (Boditech and Menarini). Therefore, we can confidently assert that AFIAS-1 and AFIAS-10 are reliable instruments for accurately and rapidly measuring 25-OH-D levels. #innovation #healthcare #technology Full Pdf link: https://lnkd.in/dSHVAyXf

Some nice work by Coombes et al, looking at ssDNA assay for product purity 🛎Overview: Coombes et al have developed a novel method using ion pair reversed phase HPLC assay for ssDNA re-generation post PCR for selective enhancement of ligands by exponential enrichment (SELEX). Source: Biomedical chromatography : BMC A liquid chromatography-tandem mass spectrometry method with vonoprazan fumarate-d4 as a stable isotope-labeled internal standard was developed and validated for the quantification of vonoprazan fumarate in human plasma. The method involved protein precipitation and gradient elution using formic acid and acetonitrile. The linear range of quantification was 0.150-60.000 ng/ml. The method was fully validated and successfully applied in a bioequivalence study of vonoprazan fumarate tablet among healthy Chinese volunteers.
