Here's how it works: - High oxygen levels (normoxia): HIF-1α is degraded, preventing the activation of pro-angiogenic genes. - Low oxygen levels (hypoxia): HIF-1α is stabilized, activating pro-angiogenic genes like VEGF, promoting angiogenesis. This balance is crucial in healthy tissues, but cancer cells exploit this mechanism to promote tumor growth and angiogenesis. "Proline is a nonessential amino acid, and its metabolism has been implicated in numerous malignancies. Together with a direct role in regulating cancer cells’ proliferation and survival, proline metabolism plays active roles in shaping the tumor microenvironment (TME). Cancer-associated fibroblasts (CAFs) display high rates of proline biosynthesis to support the production of collagen for the extracellular matrix (ECM). Indeed, impaired proline metabolism in CAFs results in reduced collagen deposition and compromises the growth and metastatic spread of cancer. Moreover, the rate of proline metabolism regulates intracellular reactive oxygen species (ROS) levels, which influence the production and release of cytokines from cancer cells, contributing toward an immune-permissive TME. Hence, targeting proline metabolism is a promising anticancer strategy that could improve patients’ outcome and response to immunotherapy." 🔔 New insight on targeted inhibition of TGF-β signaling 🔔 🔬 Brewer et al., have recently developed a bifunctional molecule named BDPIC (bromoTAG-dTAG proximity-inducing chimera). It is capable of targeted dephosphorylation of SMAD3, the key regulator of TGF-β, impeding TGF-β-induced transcriptional responses, such as the transcription of SERPINE-1 which encodes plasminogen activator inhibitor 1. The molecule, thus, has good therapeutic potential in diseases where TGF-β dysregulation has been implicated, including fibrosis and cancer. Lactate dehydrogenase A (LDHA) promotes glioblastoma progression by facilitating interaction between tumor cells and macrophages. Inhibiting LDHA disrupts this interaction, suppressing tumor growth and reducing macrophage infiltration. This review is more focused than yesterdays, specifically discussing macrophage metabolism in the context of hepatocellular carcinoma. It also highlights strategies to harness those processes therapeutically. The main goal of the study was to investigate the antitumoral effects of PRMT5 inhibition in soft-tissue sarcoma. PRMT5 is a member of the nine-member PRMT family enzymes, which serve as “writers” of post-translational modifications (PTMs) controlling oncogenic processes, including tumor intrinsic and extrinsic microenvironmental signaling pathways. Our anti-LDHD Triple A polyclonal antibody (HPA0417669) was used in western blot (WB) on sarcoma cell lines as one of the critical proteins involved in glycolysis.
