The first DMCCB literature highlight of 2024 is here! Recent research delves into the NLRP3 inflammasome's intricate functions. The study presents a novel, tricyclic NLRP3-binding scaffold discovered through high-throughput screening, evolving into NP3–562. Notably, NP3–562 demonstrates potent efficacy in human whole blood and complete inhibition of IL-1β release in a mouse acute peritonitis model at a 30 mg/kg po dose. An X-ray structure of NP3–562 bound to the NLRP3 NACHT domain reveals a unique binding mode distinct from known sulfonylurea-based inhibitors. This insight broadens our understanding of inflammasome modulation.
