Exciting news! My mentor Assoc. Prof. Rok Frlan, Ph. D. and I have recently co-authored a research paper entitled "Navigating the Chemical Space of ENR Inhibitors: A Comprehensive Analysis", which has been recently published in Antibiotics in the special issue Novel Antimicrobial Agents: Design, Synthesis and Biological Evaluation. In our study, we curated and analysed a dataset of 1412 small molecules identified as ENR inhibitors (ENR = enoyl-acyl carrier protein reductases, in particular FabI, FabK, FabV, and InhA), examining different structural variants. Using advanced cheminformatic tools, we mapped the physicochemical landscape and identified specific structural features as key determinants of bioactivity. Exciting news in blood research! Scientists have discovered enzymes from gut bacteria that remove A and B antigens from red blood cells, a breakthrough bringing us closer to universal donor blood. Led by Maher Abou Hachem, PhD, and Martin L. Olsson, PhD, the team from DTU and Lund University published their findings in nature microbiology. With ongoing research, they aim to refine these enzymes, potentially revolutionizing blood transfusion safety. 📢 New preprint from the lab in collaboration with Canan Atilgan's group. Study led by Isik Kantarcioglu. 🔬 Our study sheds light on the complex battle against antibiotic resistance, focusing on the role of the efflux protein TolC in Gram-negative bacteria. By employing a blend of experimental and computational methods, including molecular dynamics simulations and whole-gene-saturation mutagenesis, we have studied how TolC mediates the efflux of β-lactam antibiotics like oxacillin, piperacillin, and carbenicillin. Our findings reveal the critical movements and residues crucial for TolC's function but also pinpoint general and drug-specific efflux mechanisms. This deep dive into TolC's structural dynamics offers structural insights, paving the way for developing new antibiotics designed to circumvent bacterial efflux mechanisms and combat multidrug resistance. 🧬💊 #AntibioticResistance

In the last post for this week, I would like to highlight a recent study in collaboration with UCL, in which we made use of our 🔝 #SHIME platform to compare the colon-targeting efficacies of two IBD medications (i.e. #Azulfidine and #Octasa). We showed that a formulation strategy (harnessing pH + microbial release triggers) delivered more mesalazine to the colon than a prodrug strategy (sulfasalazine). We also showed that a #SHIME model that accurately simulates the GI physiology and colonic microbiota of the patient population is necessary 𝐭𝐨 𝐨𝐛𝐭𝐚𝐢𝐧 𝐫𝐞𝐥𝐞𝐯𝐚𝐧𝐭 𝐝𝐚𝐭𝐚 𝐟𝐨𝐫 𝐭𝐡𝐞 𝐭𝐚𝐫𝐠𝐞𝐭 𝐩𝐨𝐩𝐮𝐥𝐚𝐭𝐢𝐨𝐧, as in vitro models that simulate healthy individuals could result in biased conclusions about the GI performance of medications. 📄 Please follow this link to read the paper: https://lnkd.in/eSknZqgp 👏 Thanks to all co-authors!
