Researchers from Penn State shed light on the intricate regulation of cell-cell adhesion (https://lnkd.in/gKvDJJvt) . Cadherin family proteins are pivotal in maintaining epithelial and endothelial integrity. Through their investigation, they've unveiled the role of MARCH family ubiquitin ligases in cadherin degradation. They have shown MARCH2's specific impact on VE-cadherin, disrupting adherens junctions and endothelial barrier function. Interestingly, N-cadherin remains unaffected, indicating unique regulation among cadherin proteins. Learn about this non-toxic alternative to Trypan Blue ⬇️ Erythrosin B is a biosafe and non-toxic colorimetric dye that can be used as an alternative to Trypan Blue. Trypan Blue dye is commonly used in labs for cell viability, but it is also known to be carcinogenic, environmentally damaging, and cytotoxic. Our Application Scientist team conducted an analysis to compare the accuracy of Erythrosin B to Trypan Blue in measuring the viability of cell lines. The data gathered confirmed that Erythrosin B was as consistent and reliable as Trypan Blue for brightfield cell counts. Many emerging disease and cancer therapies are based on “small molecules,” compounds that, when given as drugs, induce certain therapeutic effects by binding to their targets. But knowing which small molecule will bind where is a difficult question — one not infrequently answered by sheer chance. In a research partnership with University of Pennsylvania Perelman School of Medicine's Luca Busino, Ph.D., Wistar’s Salvino lab — led by Dr. Joseph Salvino — has co-developed a new approach for systematically identifying which small molecules bind to which drug targets. Their new methodology, called TAP-DAP (Tandem-Affinity Purification for identification of Drug-Binding Proteins), not only allows researchers to systematically identify drug–protein interactions — it also indexes “off-target” interactions. By indicating where a small molecule could bind in addition to its intended target, TAP-DAP can help researchers anticipate whether a drug candidate might have either unintended toxicity or a therapeutic synergy. Struggling with media volume consistency in your cell plates? Well, it's #TechTuesday which means it's time for another technical tip from one of our scientists here at Axol to help you! Helena Raine, one of our Research Assistants at Axol that works on cellular differentiations and assays, shares her top technical tip to help you avoid media volume inconsistencies that can arise from evaporation that can take place during cellular incubation. Her top tip is to add some Phosphate-buffered saline on the outer plates beside the cells to stop evaporation in the outer well. This will help ensure that the volume in the plates remain s consistent. Media volume consistency is very important especially when performing compound screening as inconsistencies could adversely effect results. Additionally, evaporation from cell culture plates can cause culture problems since the medium components, can then reach concentrations that can become harmful to the cells. At Axol, we've spent more than a decade at the forefront of iPSC technology, adding to our bank of expertise in using iPSC-derived cells for drug discovery and research.
