According to this article from the Mayo Clinic in Medical Xpress, scientists have identified unique molecular signatures indicating dysfunction in the blood-brain barrier in Alzheimer's disease, which could lead to new methods for diagnosing and treating the condition. Led by Dr. Nilufer Ertekin-Taner, the team analyzed brain tissue from Alzheimer's patients and healthy individuals, studying thousands of cells across six brain regions. They focused on pericytes and astrocytes, key cells maintaining the blood-brain barrier, and discovered altered communication between these cells mediated by molecules VEGFA and SMAD3. Their findings suggest that elevated VEGFA levels in Alzheimer's patients reduce SMAD3 levels, impacting vascular health. Using cellular and zebrafish models, the researchers confirmed this relationship, observing decreased SMAD3 levels in brain pericytes treated with VEGFA. Higher blood SMAD3 levels correlated with less vascular damage and better outcomes in Alzheimer's disease. We're thrilled to announce a publication in Nature Portfolio - Nature Communications that highlights recent research in Alzheimer's disease detection using the #icpTOF in combination with the Teledyne Photon Machines IRIDIA laser system. The specific work features a novel peptide-coated gold nanocluster that targets connective tissue growth factor (CTGF). CTGF, an upstream regulator of ß-amyloid plaques, plays a role in Alzheimer's disease. This innovative probe shows great promise for diagnosing early-stage Alzheimer's disease before Aβ plaques form. Drug repairs systems that drain Alzheimer's-causing waste from brain The study, published in the Journal of Clinical Investigation, examined mice with craniosynostosis, a skull malformation that increases pressure inside the skull and reduces the ability to form, maintain and use the network of lymphatic vessels that help drain brain waste and plaque-laden cerebrospinal fluid. In addition, the ability of cerebrospinal fluid to perfuse into the brain and clear waste to surrounding lymphatic vessels, known as the glymphatic system, was impeded. Using a mouse model for familial Alzheimer’s disease, the addition of craniosynostosis and associated impairments to the lymphatic and glymphatic systems caused a significant increase in plaque burden in the brain. To counteract these deficits, the investigators turned their attention to a drug called Yoda1, which activates a force-sensing ion channel known as Piezo1. This drug reduced intracranial pressure and allowed newborn mice with craniosynostosis to develop and maintain normal lymphatic vasculature that supported the ability of cerebrospinal fluid to perfuse into the brain and drain its waste to lymph nodes. Researchers also found that using Yoda1 in previously untreated aged adult mice improved meningeal lymphatic and glymphatic functions, making brain waste-clearance systems that had looked old appear young again. Publication Alert! Alzheimer's & Dementia has published important research that used C2N Diagnostics’ PrecivityAD® blood test to measure study participants’ Aβ42/40 ratios and ApoE proteotypes. Are bioengineered brain tissues the key to better neurodegenerative disease treatments? That's the topic addressed in this Biomaterials review article from Mullis and Kaplan, who discussed the use of complex bioengineered models for conditions including Alzheimer's Disease and Parkinson's Disease. Our key takeaways were: • The limited complexity of simple cell cultures and the persistent translational gap from animal models means there is a pressing need for complex, human-relevant models including microfluidics-based and organoid-based bioengineered tissues • Human brain tissue is particularly complex in its structure and development. This needs to be taken into account when constructing these models. At Axol Bioscience, we're excited by the prospect of complex co-culture models and advanced platforms (including microfluidics and organoids) to better recapitulate the in vivo state • iPSC technology holds great potential for constructing patient-specific and disease-specific models.
