Sepax Unix-C SEC columns were used in this recent Janssen paper to prepare and characterize the HIV-1 Env gp140, and then used in platform MASCALE (Mass Spectrometry Enabled Conversion to Absolute Levels of ELISA Antibodies). The HIV-1 Env gp140 proteins were produced in HEK293E cells and purified with Galanthus nivalis lectin bead-based affinity chromatography. Purified proteins show largely uniform trimeric content, with occasional hexamer contributions for some clade B strains. Source: Journal of proteome research A new method using parallel reaction monitoring (PRM) and microflow LC-MS has been developed for high-throughput glycan profiling of human serum IgG subclasses. This method allows for the quantification of Fc glycopeptides and distinguishes between IgG3 and IgG4 without the need for further enrichment. The method has been shown to effectively monitor the relative levels of 13 representative glycoforms with good detection limits for individual IgG subclasses.
