dCODE Dextramer® reagents can help identify new TCR sequences for subsequent expression of functional TCRs. In our latest poster from AACR, we showcase how we identified three α-β TCR sequences of interest from dCODE Dextramer® sequencing data on ligand-specific MAIT cells generated using the BD Rhapsody™ Single-Cell Analysis System. The TCR sequences were manufactured and refolded using our TCR Solutions platform, and evaluated for α-β pairing, functionality, and specificity using SDS PAGE and an in-house developed artificial cell (beads) assay. Two of the selected TCR sequences demonstrated correct folding, specificity, and functionality, as confirmed by their recognition of the 5-OP-RU MR1 Monomer on beads.
