The recently released RevioTM instrument from Pacific Biosciences provides 8X higher throughput than the Sequel II TM enabling long read applications at unprecedented scale. However, fragmentation is typically performed through low-throughput mechanical means, and multiple micrograms of genomic DNA are needed to make a high-quality library. We have developed a PCR stop primer that can create DNA adhesion ends at will by photochemical reaction and reported it in a paper. This technology enables the automation of DNA construction. Using this method, we succeeded in synthesizing a linear lambda phage genome. This method can efficiently construct DNA sequence libraries and be applied to creating and screening mRNA sequence libraries. https://lnkd.in/dYEEehuw
