The CRISPR/Cas9 system is a method used for modifying the genomes of plants. Typically, the process of genome editing in plant genomes using CRISPR/Cas9 involves four distinct phases. Initially, users employ several web-based tools for sgRNA production to create sgRNAs, specifically targeting a gene of interest. These tools prompt users to input the location, name, or DNA sequence of each pertinent gene, together with details about the species under consideration. Plants exhibit increased expression of sgRNA in the presence of short nuclear RNA promoters, specifically U3 and U6. To enhance the accuracy of computational sgRNA identification, it is necessary to conduct extensive data collection and in-depth investigations on sgRNA in plant cells. In the second stage, it is necessary to utilize a transitory transformation method, such as hairy root or protoplast transformation using CRISPR, in order to confirm the functionality of sgRNA before employing it for genetic manipulation. In the third stage, the process entails the steady incorporation of sgRNA and Cas9 into the genetic material of the plant, together with the introduction of a genome editing construct into plant cells. This is often achieved using methods such as particle bombardment or Agrobacterium-mediated transformation.
