We are thrilled to announce the release of our latest research pre-print on a novel strategy for calculating absolute binding free energies (ABFEs) in protein-ligand systems using simple geometric collective variables and OneOPES, the enhanced sampling scheme we recently developed in Francesco Luigi Gervasio's lab. Tested on BRD4 and Hsp90 with 17 diverse ligands, our method predicts binding affinities with a mean error within 1 kcal/mol of experimental values. This approach requires no system-specific tailoring and effectively samples binding modes, aligning with experimental structures. Here is the link: https://lnkd.in/dUVxZmGr

The effectiveness of many antibiotics depends on their uptake by bacterial cells. Especially when it comes to Gram- bacteria, the presence of an additional outer membrane makes the internalization of antibiotics even more complicate, as this impermeable barrier could prevent many molecules from reaching their targets. Despite the importance of this process, surprisingly little is known on the topic so far. In our recent work, we investigate how Escherichia coli translocates fosfomycin through its outer membrane, and we highlight the importance of the experimental conditions to fully understand the translocating role of different porins. In particular, we show the physiological role of OmpF, OmpC and LamB. I am pleased to share with you my recent publication in Angewandte Chemie - "Rhodium-Catalyzed Asymmetric Macrocyclization towards Crown Ethers Using Hydroamination of Bis(allenes)". Chiral crown ethers (CEs) exhibit strong asymmetric induction in phase transfer catalysis, supramolecular catalysis and molecular recognition processes. In this study, a practical catalytic asymmetric macrocyclization method is described for the gram-scale synthesis of a class of chiral CEs using Rh-catalyzed hydroamination of bis(allenes) with diamines. Using this approach, a wide range of chiral CEs with ring sizes ranging from 12 to 36 were successfully prepared with high enantioselectivity. This project was accomplished in the Breit Group.
