In a new study, researchers at Weill Cornell Medicine, including CRI Lloyd J. Old STAR Dr. Gregory Sonnenberg, identified a previously unknown link between two key pathways that regulate the immune system in mammals. "Until now, most research on CTLA-4 focused on T cells, another type of immune cell. IgA autoantibodies in Alzheimer Disease✨✨ While microglia, the resident immune cells of the central nervous system, play a crucial role in AD, recent evidence suggests the infiltration of cerebral vessels and parenchyma by peripheral immune cells, including neutrophils, T lymphocytes, B lymphocytes, NK cells, and monocytes in AD. Obviously, if peripheral immune cells can use the breaches in the BBB, IgA autoantibodies are also able to penetrate to the lesions. More evidence of the involvement of neutrophils are source of MPO activity is the subject of current JJP Biologics scientific post. Because due to breaches in the BBB, next to CD89 expressing and resident microglia, CD89-expressing peripheral neutrophils and monocytes get access to the CNS. The current manuscript shows data in the human AD brain suggesting that neutrophils accumulate, and are associated with blood vessels in AD where they may contribute to vascular stalling and BBB leakage. It was also shown that neutrophils and NETs are the key source of MPO in the brain during AD which may represent an important mechanism through which BBB inflammation influences oxidative stress in AD. https://lnkd.in/g92AWPUV Since IgA autoantibody deposits are found in AD brain, and the observation that these are of the IgA isotype is not coincidental. These IgA autoantibodies could, when the bind their target in brain, provide an IgA matrix to myeloid cells like neutrophils, monocytes and also resident microglia through cross-linking of the the CD89 receptor resulting in activation of MPO and consequently resulting in severe tissue damage. The oxidative force of MPO is one of the causes of the tissue damage. For this reason, JJP Biologics has developed JJP-1212, which completely blocks the potent effector functions like MPO activation of these IgA auto-antibodies.
