Chemists from MIT have developed a novel system to synthesize complex molecules known as oligocyclotryptamines. Oligocyclotryptamines are complex compounds originally isolated from plants that demonstrate potential for use in an array of pharmaceutical products including antibiotics, analgesics, or cancer drugs. While promising, only small quantities of oligocyclotryptamines are naturally available. To overcome this lack of availability, MIT chemists developed a method, published in the Journal of the American Chemical Society, to add tryptamine-derived components one at a time to molecules. Over the next three years, we aim to develop scalable, efficient, and sustainable techniques for producing drug intermediates with enhanced solubility, bioavailability and controlled solid form. 🔬 Drug development is costly and risky. Late Stage Saturation (LSS) vs Late Stage Functionalization (LSF) Converting aromatic molecules into saturated analogs with a higher fraction of sp3 carbons holds promise for enhancing their medicinal properties. This study demonstrates the feasibility of saturating aromatic and heteroaromatic drugs through mild rhodium-catalyzed hydrogenation, acid-mediated reduction, or photocatalyzed-hydrogenation techniques. By transforming sp2 carbon atoms into sp3 carbon atoms, can create saturated molecules with improved medicinal characteristics. Notably, the rhodium-catalyzed approach accommodates traces of dimethyl sulfoxide (DMSO) or water, enabling the reformulation of pharmaceutical compound collections stored in wet DMSO for chemical synthesis. Exemplified through the late-stage saturation (LSS) of 768 complex and densely functionalized small-molecule drugs, showcasing the transformative potential of these techniques in drug development.
