Using nucleases with high off-target editing… Everything is not 'fine' when you have high off-target editing caused by inefficient nucleases. Enhancing bystander effects through EV-mRNA cargo in AAV gene replacement therapy: in their latest study, Tierra Bobo at University of North Carolina at Chapel Hill and collaborators created a rAAV vector with a 25 nt EV-mRNA zip code sequence linked to human HGSNAT cDNA to enhance bystander effects, aiming to develop an effective rAAV9 gene replacement therapy for treating MPS IIIC https://lnkd.in/egwA3d4K These results strongly support the therapeutic potential of this novel rAAV gene therapy for MPS IIIC. The authors believe this approach could broadly target diseases involving non-secreted proteins or transcription factors, addressing safety concerns related to high vector doses.
