BRETing success: Fine tuning whole cell NanoBRET assays. I am excited to share some recent work I’ve been doing at Charnwood Discovery. I set out to assess target engagement of ATP-competitive binders to a family of three homologous ATPase proteins via NanoBRET assays. The study led to the development of three bespoke whole cell target engagement assays despite initial challenges. It also provided further understanding of the relative endogenous ATP affinities of the three ATPases to inform their likely druggability.
