A new paper in Lab Animal from Dr Ben Davies describes a method to reduce and refine the use of mice when producing genetically modified mouse models, using sterile hybrid male mice generated by breeding two wild-type mouse strains: https://lnkd.in/dNFUsHZJ Before modified embryos can be implanted, recipient mice must be mated with sterile males to create a pseudopregnancy for embryo transplant. Male mice either undergo surgical vasectomy under anaesthesia or require extensive breeding programmes to generate sterility. In collaboration with the Mary Lyon Centre at MRC Harwell and The University of Manchester Faculty of Biology, Medicine and Health, funded by an NC3Rs skills and knowledge transfer award, Ben shows using sterile males, bred from C57BL/6J and Mus spretus strains, to prepare pseudopregnancy transfer recipients results in a similar pregnancy rate, birth rate and litter size compared to traditional methods. We've just published the results of an N3CRs-funded study in Lab Animal. We build on our idea of using naturally sterile hybrid mice to produce embryo transfer recipients - we confirm the suitability of a more commonly available hybrid at 3 different facilities (Oxford, Manchester and Harwell). It's great to see the 3Rs value of our approach being validated by independent teams. https://lnkd.in/dfEecBnQ

#customerpublicationweekly Air #pollution has been shown to be harmful to the #reproductive system. A recent publication in Cell Discovery shows that, strikingly, the paternal exposure to PM2.5 could lead to hypogonadism in the male offspring. Specific small RNAs seem to play a critical role in this trans-generational effect. #singlecell #RNAseq with Singleron's #GEXSCOPE product identified genes whose expression patterns are altered in Leydig and Sertoli cells in the testis of the male offspring. https://lnkd.in/dYYRB_rv

🦈 In the wild, the Greenland shark can live to an impressive age of up to 392 ± 120 years, making it the longest-lived vertebrate. One might conclude that studying the Greenland shark would be the best approach to understanding ageing. However, this is only partially true. 🐁 Despite its impressive lifespan, the Greenland shark is not a good model animal for studying ageing. Several factors make the humble field mouse a much more practical research tool. Aside from the very much obvious considerations related to housing and feeding, studying ageing in a short-lived organism provides relatively rapid access to data. At the Biotest Facility, we are equipped and qualified to conduct research in biogerontology using mouse models. 📧 If you plan to undertake studies of drug efficacy, pharmacodynamics, pharmacokinetics, or biomarkers in aged mice, contact us at info@biotestfacility.com for further information and support. #Aging #AnimalResearch #Preclinical #DrugDevelopment #LifeScience

The journey of drug development is exploratory and variable. Target identification and validation stages encompass a multitude of experiments and approaches to ensure confidence in the target’s biological and therapeutic potential. In our most recent blog post, we provide a brief overview of how histopathology can be used as a quick and relatively efficient way to obtain initial data during the target identification and validation phase.
