By leveraging cutting-edge proteomic technologies and mass spectrometry workflows developed by Momentum, Bowers and colleagues were able to identify and characterize several covalent cysteine modifiers active against cancer-driving helicases. Their research demonstrates the power of mass spec covalent binding screening, modification mapping, and occupancy calculations in taking a project from screening to hit characterization with unmatched efficiency. Nice paper by Denali Therapeutics, taking advantage of ATUM's orthogonal transposons Leap-In 1 and Leap-In 2. They compare co-transfection vs. supertransfection (transfection on a previously transfected pool) of Gene-of-Interest to increase titer in stable CHO. Show high expression efficiency with similar expression levels independent of transfection method. Supertransfection using orthogonal transposases is a valuable strategy for increasing titers or for overexpressing helper genes to improve the expression and/or product quality of existing pools, saving significant time and resources.

When discussing lipid nanoparticles (LNPs), we often focus on the drug payload, but the lipid components, their structure, and their impact on gene delivery are equally crucial. Cholesterol, the second most abundant component after ionizable lipids, plays a key role. Adding cholesterol improves delivery efficiency and enhances LNPs' ability to fuse with cell membranes and release their contents. To support researchers in their formulation and process scale-up studies, Corden Pharma - A Full-Service CDMO created #LNP starter kits. These kits provide up to 15 mL of a complete lipid mixture and offer a variety of lipid components, including cholesterol or its analogs. Evaluating #cholesterolanalogs is essential; for instance, researchers have studied LNPs containing naturally occurring phytosterols like β-sitosterol, stigmasterol, and stigmastanol. They discovered that incorporating these cholesterol analogs significantly changes the LNPs' structure and #improves #nucleicacid #delivery (Eygeris et al. Deconvoluting Lipid Nanoparticle Structure for Messenger RNA Delivery. Nano Lett. 2020 Jun 10;20(6):4543-4549.)
