Ian Hay at Factor Bioscience Inc. gave an incredible presentation showcasing research that could revolutionize the treatment of solid tumours. At the heart of this innovation is a novel method for producing macrophages from induced pluripotent stem cells (iPSCs). Hay described an optimized 28-day protocol that is both feeder-free and xeno-free, making it amenable to GMP-compliant production. Thanks for sharing, Ian!     🧫🔬 𝐈𝐧𝐯𝐞𝐬𝐭𝐢𝐠𝐚𝐭𝐢𝐧𝐠 𝐏𝐥𝐮𝐫𝐢𝐩𝐨𝐭𝐞𝐧𝐜𝐲 𝐌𝐚𝐫𝐤𝐞𝐫 𝐄𝐱𝐩𝐫𝐞𝐬𝐬𝐢𝐨𝐧 𝐔𝐧𝐝𝐞𝐫 𝐃𝐢𝐟𝐟𝐞𝐫𝐞𝐧𝐭 𝐆𝐫𝐨𝐰𝐭𝐡 𝐅𝐚𝐜𝐭𝐨𝐫 𝐂𝐨𝐧𝐜𝐞𝐧𝐭𝐫𝐚𝐭𝐢𝐨𝐧𝐬 𝐚𝐧𝐝 𝐅𝐞𝐞𝐝𝐢𝐧𝐠 𝐂𝐨𝐧𝐝𝐢𝐭𝐢𝐨𝐧𝐬   Following the cell morphology and proliferation analysis examined in the past few posts, we went on to analyse pluripotency.   The effects and capacity of FGF-2 TOP® to maintain high-quality iPSC cultures under different concentrations and media feeding regimes were assessed by determining the expression of key markers of pluripotency:   OCT4 NANOG SSEA5 SSEA4     The results show that FGF-2 TOP® presents a superior performance for culturing iPSCs and allows you to reduce media feeding – including weekend-free schedules - without compromising the quality and pluripotency of the cells in culture.   The details of the flow cytometry approach and different culture conditions can be found in the study - https://lnkd.in/eM6Hm7kR

    📈 𝐢𝐏𝐒𝐂 𝐏𝐫𝐨𝐥𝐢𝐟𝐞𝐫𝐚𝐭𝐢𝐨𝐧 𝐀𝐧𝐚𝐥𝐲𝐬𝐢𝐬 📈 We assessed the growth rates of iPSCs under different concentrations of both 'wild-type' human recombinant FGF-2 and recombinant FGF-2 TOP®(Thermostability Optimized) using cell proliferation assays. After 5 days in culture, the lower concentration (50 ng/mL) promoted an average amplification rate of 73% for human recombinant FGF-2, while for recombinant FGF-2 TOP® the amplification rate corresponded to 82%. The effects of adding a higher concentration of 100 ng/mL resulted in an amplification rate of 83% and 85% for human recombinant FGF-2, and FGF- 2 TOP® respectively. Further analysis with every other day (EOD) media replacement, demonstrated the capacity of FGF-2 TOP® to keep high proliferation rates of iPSCs even under reduced feeding schedules. Conclusion – Thanks to the thermal stability of FGF-2 TOP®, less FGF-2 is needed for comparable proliferation rates, contributing to lower costs and more flexible culture operations.     📖 New Study: 𝐄𝐯𝐚𝐥𝐮𝐚𝐭𝐢𝐧𝐠 𝐭𝐡𝐞 𝐮𝐬𝐞 𝐨𝐟 𝐓𝐡𝐞𝐫𝐦𝐨𝐬𝐭𝐚𝐛𝐥𝐞 𝐅𝐆𝐅-2 (𝐅𝐆𝐅-2 𝐓𝐎𝐏®) 𝐟𝐨𝐫 𝐨𝐩𝐭𝐢𝐦𝐢𝐳𝐞𝐝 𝐢𝐏𝐒𝐂 𝐂𝐮𝐥𝐭𝐮𝐫𝐞 𝐏𝐫𝐨𝐭𝐨𝐜𝐨𝐥𝐬   iPSC culture requires FGF-2 to maintain pluripotency, but it comes at a cost.   The FGF-2 protein is inherently unstable - <10 hours half-life! - leading to difficulties sustaining signalling cues for pluripotency.   The result: daily media changes, which are a constant barrier for researchers and therapy developers.   🧫 So, can we enhance the efficiency and cost-effectiveness of iPSC culture techniques?   We think so.   🌡️ Our thermostable version of FGF-2 comes with an enhanced half-life (> 7 days at 37ºC), developed by computer-assisted engineering, licensed from Enantis. Key Highlights: Processing Aid: Our FGF2-STAB® protein is classified as a processing aid under Regulation (EC) No 1333/2008. This confirms that while there may be technically unavoidable residues, they pose no health risk. This milestone underscores the crucial role our protein plays in improving the efficiency and quality of cultured meat production. Non-GMO: Our FGF2-STAB® protein is not considered a GMO. The only applicable status would be “produced with” GMO which is not relevant for processing aids. FGF2-STAB® can be used in cultured meat products across the European market without the regulatory challenges associated with GMOs, as per Regulation (EC) No 1829/2003.
