New research led by Montefiore Einstein Comprehensive Cancer Center’s Roberto Alejandro A. Sica, MD, and colleagues explores allogeneic chimeric antigen receptor (CAR) T-cell therapy as a potential treatment for relapsed/refractory T-cell lymphomas. Dr. Sica is an Attending Physician at Montefiore Einstein Comprehensive Cancer Center and Assistant Professor, Medicine at Albert Einstein College of Medicine. The study, published in The Lancet Oncology on November 28, 2024, evaluates CTX130, an allogeneic CAR T-cell therapy targeting CD70—a therapy that appears to have some promise. A next-generation therapy, CTX131, is now being studied at Montefiore Einstein Comprehensive Cancer Center. The lack of effective treatments for relapsed or refractory T-cell lymphomas has meant poor survival rates for patients. Research has suggested that allogenic CAR T-cell therapy targeting CD70, a cell-surface protein overexpressed in T-cell lymphomas, could be a promising approach. In a study published online on November 28, 2024, in The Lancet Oncology, Roberto Alejandro Sica, M.D., and colleagues report the results of the first clinical trial to evaluate this therapy. In the phase 1 trial, conducted at centers in the United States, Australia, and Canada, the CAR T-cell therapy, known as CTX130, was administered to 39 people previously treated for T-cell lymphomas. Forty six percent of patients responded to the therapy, with 6 achieving complete responses, and with higher doses yielding better response rates. A variety of serious but manageable side effects occurred. The results show that this type of CAR T-cell therapy is safe and effective for a subset of patients with T-cell lymphomas. A newer-generation CAR T-cell therapy, called CTX131, is now being studied at several medical centers including Montefiore Medical Center. Osteosarcoma is the most common malignant childhood bone tumor, with deaths often resulting from the spread of the tumor to other parts of the body. In a research article published in the journal Molecular Cancer Therapeutics in January 2025, Montefiore Einstein Comprehensive Cancer Center’s David Loeb, MD, PhD, and team elaborate on a promising drug target for osteosarcoma called DKK-1, which is a protein found in bone, prostate, and other organs potentially involved in transforming cancer cells back into normal bone cells. Finding a Novel Strategy Against Osteosarcoma Osteosarcoma (OS) is the most common malignant childhood bone tumor. Most osteosarcoma deaths result not from the primary tumor but rather from metastatic disease. Decades of clinical trials have failed to improve the poor survival rates of patients with metastatic OS. In research published online on January 9 in Molecular Cancer Therapeutics, David Loeb, M.D., Ph.D., and colleagues showed that DKK-1—a protein found in bone, prostate, and other organs—is a promising target for OS therapy. DKK-1 is known to inhibit the Wnt signaling pathway, which has been implicated in osteosarcoma pathogenesis. The researchers found that a small-molecule inhibitor of DKK-1 activated Wnt signaling, causing reduced proliferation of OS cell lines and—in OS patient-derived mouse xenografts—inhibition of metastasis not by killing cancer cells but by causing them to differentiate into normal bone cells. The results justify developing therapies for OS metastasis that work by targeting DKK-1 or by manipulating Wnt signaling in other ways. Dr. Loeb is professor of pediatrics and of developmental & molecular biology and the Linda and Earle Altman Faculty Scholar in Cancer Research at Einstein.
