🎉 Excited to share our latest publication in the Journal of Diagnostics, titled "Advancing Evidence Generation for ctDNA: Lessons Learned from A Multi-Assay Study of Baseline ctDNA Levels Across Cancer Types and Stages," led by Brittany Avin McKelvey and Friends of Cancer Research. This work evaluates ctDNA levels prior to cancer treatment, facilitating evidence generation opportunities to support the use of ctDNA as a biomarker for clinical response. At ASCO 2023, we had the opportunity to showcase the results of our remarkable PHERGain study, presented by Dr. Javier Cortés Castán. The study focuses on evaluating strategies for de-escalating chemotherapy in HER2-positive early breast cancer. The outcomes, unveiled last June, have now earned their place in The Lancet, the world's preeminent academic journal, underscoring the magnitude of our achievements. The #PHERGain trial has demonstrated promising results in the treatment of HER2-positive early breast cancer. Through its adaptive treatment design, this study has paved the way for a chemotherapy-free approach, reducing the burden of toxic treatments for patients. It was a spring day, with the warm sun streaming into the room, when I received a call from Toni (Dr. Llombart-Cussac) to discuss the design of a new study, with a dynamic approach. I sat down, mindful of my growing belly. Further discussions followed with José (Dr. Perez) and Javier (Dr. Cortes), along with the dedication of many and the perseverance of countless patients and their families. And now, with my son Giovanni celebrating his 10th birthday, I am deeply moved to share with you the publication of the survival data from PHERGain, featured in The Lancet. The PHERGain study is notable for its unique design, leveraging both on treatment PET scans and pathological findings to guide physician decisions regarding chemotherapy prescription alongside anti-HER2 therapy. Today's published results demonstrate that an impressive 99% of PET responder patients who achieved also a pathological complete response (pCR) remained free of breast cancer relapse at 3 years after surgery without receiving chemotherapy at any point during their treatment trajectory. This breakthrough holds significant implications, sparing a segment of patients (those with both PET response and pCR) from chemotherapy-related toxicities, and affirming the optimization of treatment strategies while ensuring favorable long-term patient outcomes. Now, we must redouble our efforts in researching new biomarkers and utilize the innovative PHERGain study design as a roadmap to extend its positive outcomes to a broader spectrum of women.
