The NBCA Journal Club highlights groundbreaking research in thrombosis and thrombophilia and features a manuscript author as part of the discussion. Our next session convenes June 18, 2024 from 2-3 p.m. ET to discuss, Antithrombin Therapy: Current State and Future Outlook, published in Clinical and Applied Thrombosis/Hemostasis in 2023. The paper’s author, George M. Rodgers, III, MD, PhD, Professor of Medicine in the Division of Hematology and Hematologic Malignancies at the University of Utah School of Medicine, will join us to discuss his research. The session’s learning objectives: - Understand the genetic and acquired factors contributing to antithrombin (AT) deficiency and its implications. - Explore current treatment strategies for managing AT deficiency. - Identify specific patient populations and use cases where AT therapy may be considered and evaluate its potential benefits. - Investigate advancements and future directions in research, diagnosis, and treatment approaches for AT deficiency to enhance clinical outcomes and patient care. I am excited to announce you to check out our publication. https://lnkd.in/dcZqMs_t TITLE The PI3K/Akt signaling axis in Alzheimer’s disease: a valuable target to stimulate or suppress? ABSTRACT Among the long list of age-related complications, Alzheimer’s disease (AD) has the most dreadful impact on the quality of life due to its devastating effects on memory and cognitive abilities. Although a plausible correlation between the phosphatidylinositol 3-kinase (PI3K) signaling and different processes involved in neurodegeneration has been evidenced, few articles reviewed the task. The current review aims to unravel the mechanisms by which the PI3K pathway plays pro-survival roles in normal conditions, and also to discuss the original data obtained from international research laboratories on this topic. Responses to questions on how alterations of the PI3K/Akt signaling pathway affect Tau phosphorylation and the amyloid cascade are given. In addition, we provide a general overview of the association between oxidative stress, neuroinflammation, alterations of insulin signaling, and altered autophagy with aberrant activation of this axis in the AD brain. The last section provides a special focus on the therapeutic possibility of the PI3K/Akt/mTOR modulators, either categorized as chemicals or herbals, in AD. In conclusion, determining the correct timing for the administration of the drugs seems to be one of the most important factors in the success of these agents. Also, the role of the PI3K/Akt signaling axis in the progression or repression of AD widely depends on the context of the cells; generally speaking, while PI3K/Akt activation in neurons and neural stem cells is favorable, its activation in microglia cells may be harmful.

Caroline Mohammed (laboratory technician, University of North Carolina, Chapel Hill) and Rohan Rao (medical student, University of Cincinnati) recently published a review of the use of plant extracts in medicine to modify ion channels in cancer symptomatology and treatment. This is Caroline's first attempt at writing a review review. Ion channels have become increasingly important in cancer treatment, and plant extracts have properties that can modulate these channels. 📚 Thrilled to announce the publication of our latest literature review article! 📝 Titled "Extracellular vesicles: the key to unlocking mechanisms of age-related vascular disease?", this piece explores the intricate relationship between aging and the development of two prevalent pathologies: vascular calcification and amyloidosis. As we age, our bodies face challenges, and two of the most prevalent are vascular calcification and amyloidosis. These conditions involve the accumulation of toxic aggregates in the vessel extracellular matrix (ECM), impacting ECM integrity and driving vascular stiffening. What's intriguing is the commonality in the genesis of these pathologies, particularly their reliance on a nucleus to mediate their formation. In the article, we delve into emerging evidence suggesting that changes in ECM composition associated with cellular senescence, along with the role of extracellular vesicles (EVs) and vascular smooth muscle cells (VSMCs), play pivotal roles in the development of both calcification and amyloidosis. Protein aggregation pathologies in #Parkinson #PD #DLB #MSA and other forms of #neurodegeneration and #dementia are diverse and complex. This is critical because these pathologies are i) often considered one and the same, ii) not distinguishable using conventional pathological markers -- and -- iii) many biological and drug discovery screens thus do not distinguish among them. Phenotypic screens are only as good as the phenotype one is screening against! In this work we present a robust, tractable and screenable suite of human and patient-specific #stemcell #IPS #IPSC models now puts this complexity within reach in the laboratory dish.
