Enzymes can’t tell artificial DNA from the real thing. Researchers at the UC San Diego have made progress in expanding the genetic alphabet by demonstrating that RNA polymerase, a key enzyme involved in protein synthesis, can recognize and transcribe artificial base pairs. The study uses a version of the genetic alphabet called the Artificially Expanded Genetic Information System (AEGIS), incorporating two new base pairs. The findings suggest that synthetic base pairs could be used in the transcription process similarly to natural base pairs, potentially opening up new possibilities for synthetic biology and the design of custom proteins for therapeutic applications. 🌹 Happy Valentine’s day 🌹 🌹 Researchers have developed an efficient CRISPR/Cas9 platform for precise genome editing in roses, a task previously challenging due to the plant's genomic complexity and lack of transformation technology. 🌹 Using suspension cells, they screened for highly efficient single-guide RNAs (sgRNAs) and optimized the CRISPR/Cas9 system by modifying the promoter to drive sgRNA expression. 🌹 Successful editing of RhEIN2, a gene essential in the ethylene signaling pathway, resulted in ethylene insensitivity. 🌹 The system, tested on various genes including RhPDS, RhCAT, RhJAZ, and RhH6, showed higher mutagenesis efficiency with the pYLCRISPR/Cas9Pubi-H-A construct. 🌹 Suspension cell culture facilitated sgRNA evaluation before stable transformation, streamlining the process. 🌹 Editing RhEIN2 led to reduced gene expression and compromised ethylene response, demonstrating the platform's potential for functional genomics and molecular breeding in roses. 🌹 Additionally, it opens avenues for synthetic biology and transgenic technology applications, such as producing rose-derived fragrance and cosmetic raw materials.
