In a correspondence letter we wrote for the New England Journal of Medicine (#NEJM), we suggested that testing for POLE, and even better, checking for POLE-associated mutational signatures for example by whole-genome sequencing, could help understand why some endometrial patients with mismatch repair proficiency respond to immunotherapies. I advocate that every patient with an aggressive endometrial tumor should have whole-genome sequencing.
