    📌 The DNA Damage Response (DDR) pathway preserves genomic integrity by detecting and repairing DNA damage, ensuring proper replication, cell cycle progression, and apoptosis. DDR dysfunction contributes often to cancer by promoting genomic instability and uncontrolled growth. Given its central role in cell cycle progression and cell proliferation, the DDR pathway has become a significant target for anti-cancer therapeutics.  📌 Targeting DDR with inhibitors enhances cancer cell sensitivity to chemotherapy and radiotherapy, often leveraging synthetic lethality, as seen with PARP inhibitors in BRCA-mutant cancers. 📍 Promega Corporation offers a variety of assays and technologies to facilitate PARP/DDR pathway drug discovery.     The Efficiency of Gene Delivery through AAV can be challenged by preexisting patient immunity, dosage, tissue tropism. Neutralizing antibodies (NAbs) in serum can interfere with gene therapy, particularly when using viral vectors like AAV. Their removal require specialized procedures and have limitations. Detecting and quantifying these antibodies is therefore crucial for patient screening, monitoring immune responses and patient eligibility. A cell-based reporter such as Promega’s Transduction Inhibition Assay allows to measure AAV uptake of 11 AAV serotypes and it is suitable to determine tissue tropism in vivo. All of this with a simple and short (4-24 hours incubation) workflow. The assay is useful for 📌 quantifying Nab titer in human serum samples 📌 Early development, pre-clinical and clinical studies 📌 Screening AAV capsid variants to evade Nab 📌 Monitor Nab levels to optimize redosing and is suitable for early development, pre-clinical and clinical studies.
