A fantastic day at FIGON Dutch Medicines Days 2024! The best thing? Catching up with Miriam Amiguetti Grande, JPP's former intern now working at Hyphen Projects, the conference organizer. Here’s what we learned 👇🏼 The conference started with Rob Nelissen from Organon, pointing out the need to invest in people to close the pharma industry's expertise gap. He introduced Pharma Delta, a new alliance bringing together companies, universities, and government bodies to boost global health. We then enjoyed a panel discussion with Prof. Ilze Bot from Leiden Academic Centre for Drug Research (LACDR), Natasja Wijling (Co-chair NVOO), Prof. Patrick Rensen (Leiden University Medical Center), and Dr. Karin Conde-Knape (Novo Nordisk), about the obesity pandemic. Why call it a pandemic? Because 16% of the global population was affected in 2022, with similar numbers in previous years. The speakers explained that tackling this complex disease requires a combination of treatments, education, and lifestyle changes. During the first session, Challenges and Strategies in the Development of Anti-Cancer Nanomedicine, Esmee Koedoot from VitroScan presented their ex vivo tumor testing, which is improving predictions for cancer therapies, and Willem Mulder from Eindhoven University of Technology talked about nanotechnologies using apolipoproteins as delivery platforms of drugs to immune cells. In the second session we attended, "Treatment of Brain Tumors: New Windows of Opportunities", the speakers focused on the challenges of the blood-brain barrier (BBB) in treating brain tumors and the innovative solutions to tackle them! During their discussion on stage, Elizabeth (Liesbeth) De Lange from LACDR and Dr. Dannis van Vuurden from Prinses Máxima Centrum voor kinderoncologie (PMC) highlighted the use of focused ultrasound and microbubbles for BBB opening, allowing better drug delivery to tumours. They shared promising studies showing how this method can improve brain cancer treatment. Liska Scheffers from PMC started her presentation by saying: "a petri dish is not a patient," stressing the importance of pharmacokinetics in drug discovery. She presented their work on gemcitabine, a nucleoside analogue with low plasma protein binding, which helps its better penetration through membranes like the BBB. Their studies at PMC have shown promising results. The conference closed with a take-home message by Erik (H.W.) Frijlink from the University of Groningen: technology development needs to start early and stay involved throughout the whole R&D process to simplify active compounds, explore new delivery methods like inhalation, and improve clinical results. Patricio de Boom from Microlab | Offices, Maker Space, Event Venue & Coworking gave an insightful presentation about the facilities and resources they offer to startups and researchers. From state-of-the-art labs to specialized equipment and a network of industry experts, #Microlab is a valuable asset to the entrepreneurial ecosystem. Farshida Zafar’s from the Erasmus Centre for Entrepreneurship kicked off the evening with some fun icebreakers, creating a relaxed and engaging atmosphere. Animal testing in drug development, is it the future? Last week, we attended the Animal-Free Tech Talk at Leiden Bio Science Park, where experts got together to explore the future of drug development without animal testing. Did you know that nearly half a million animal tests were done in 2022, but 90% of drugs that pass animal studies still fail in human clinical trials? Shocking, right? That's what Debby Weijers from Proefdiervrij highlighted. André Wolterbeek from Venn Life Sciences - part of hVIVO plc pointed out that although non-clinical testing helps to minimize adverse effects and provide data for human trials, the high failure rate in later-stage trials shows the need for alternative methods to preclinical animal testing. Here are the key highlights: StemX Bio - Elena Naumovska presented their 3D pancreatic models that mimic human pancreas complexity, providing a faster, cheaper and more predictive alternative to animal models. ExCulture – Aisling Foley talked about their animal-free heparin substitute that could disrupt the market by reducing the need for animal sources in producing this crucial blood thinner. FyBrix – Paul Kouwer introduced their synthetic matrix technology that replaces mouse-derived Matrigel in cell cultures. It’s more human-relevant and helps cut down on animal testing. chiron – Dustin Dopsa shared their "organ-on-a-chip" technology, which mimics the human joint to test how cells respond to new drugs. Leiden Academic Centre for Drug Research - Lukas Wijaya explained how their kidney organoids are designed to better mimic human kidneys responses towards toxins and improve the current in vitro models. Peregrion - Wouter Vaes highlighted their use of accelerator mass spectrometry to spot toxic metabolites early, helping to reduce animal testing by relying on human data. Last Thursday, we dived into the exciting world of AI-powered drug development at the 3rd Leiden Drug Development Conference held at the ECC Leiden. This event, organized by Johnson & Johnson, TNO, Leiden University, and Leiden University Medical Center (with support from Hyphen Projects and Marije Stofregen), explored how #AI is changing everything from drug discovery to manufacturing and healthcare! Here's a taste of what the conference offered: AI: The Future of Drug Development? This edition focused on the transformative potential of AI across the entire drug development pipeline. We attended parallel sessions where top academics and industry leaders shared real-world applications of AI in various areas: -Drug Discovery: How AI helps us identify promising drug candidates. -Care Optimization: Using AI to personalize treatment plans for patients. -Drug Safety Assessment: AI tools that help predict potential side effects. We also had the opportunity to connect with some truly inspiring speakers: Gerard JP Van Westen Showed how AI is changing drug discovery by using vast amounts of medical data to predict drug-protein interactions. Herman van Vlijmen: His expertise provided valuable insights into AI's role in drug design and discovery. Stef van Grieken: Shared his exciting perspective on how AI is transforming manufacturing and healthcare. Simon Dalmolen Offered valuable insights on data sharing and standardization to move the field forward. The conference wasn't just about talks. We also learned about DataChaperone: A platform making complex AI models accessible to a wider audience. A Deep Dive into PKPV and AI: Rob van Wijk's presentation highlighted the potential of combining pharmacokinetic-pharmacodynamic (PKPV) modeling with AI. He discussed how this can provide deeper insights into drug mechanisms, enable personalized treatment, and accelerate the drug discovery process. He also addressed challenges like data privacy and proposed using joint distribution functions to create shareable virtual patient populations. Scaling up your business can be tricky, right? Yesterday, we attended the Biotech Scale-up event organized by Planet B.io in collaboration with hollandbio and it did not disappoint! Held at the TU Delft Hortus Botanicus, a one-of-a-kind location, the event brought together four top pilot facilities to showcase their expertise, equipment, and how they can help startups overcome the challenges of scaling biobased processes. From Mickel Jansen’s insights into bio-process development at DSM, to Hendrik Waegeman of Bio Base Europe Pilot Plant explaining how to turn grams into tonnes, the presentations were packed with practical knowledge for startups looking to scale their operations. The session continued with Martijn Bekker’s deep dive into reproducible scaling at Wageningen Food & Biobased Research, Menno Rus of Plant One Rotterdam BV discussing the critical infrastructure needed for startup success, and ended with Robyn Eijlander of NIZO sharing strategies for avoiding the “valley of death” in upscaling. Not only did this event give insights for startups looking to take their operations to the next level, but we also celebrated HollandBio’s 10-year anniversary, marking a decade of biotech innovation!
