Questions discussed will be: - What shaking parameters should I choose for my microplates cultures? - Reproducibility issues in shaken culture results, what are causes? - The scale-up from shake flask into stirred vessel, what's the issue? - Baffles, working volume, shaking diameter, stopper type, well geometry, why and how does it influence shake flask cultures? - How can I simulate a large-scale fed-batch process on a small-scale? For instance, on the attached poster you can find scientific data of the comparison of two different K. phaffii strains which were grown in batch culture on glucose, glycerol, and methanol as sole carbon source in shake flasks. Two of the key enzymes of the methanol utilization pathway (Mut) are the alcohol oxidases 1 and 2. In this study, a Mut+ strain expressing both, AOX1 and AOX2, and a MutS (Methanol utilization slow) strain only expressing AOX2 were compared. 𝐊𝐮𝐡𝐧𝐞𝐫 𝐅𝐞𝐞𝐝𝐢𝐧𝐠 𝐓𝐞𝐜𝐡𝐧𝐨𝐥𝐨𝐠𝐲 𝐞𝐱𝐩𝐚𝐧𝐝𝐬 𝐰𝐢𝐭𝐡 𝐧𝐞𝐰 𝐩𝐫𝐨𝐝𝐮𝐜𝐭: 𝐬𝐮𝐜𝐫𝐨𝐬𝐞 𝐟𝐞𝐞𝐝𝐢𝐧𝐠 𝐟𝐨𝐫 𝐩𝐥𝐚𝐧𝐭 𝐜𝐞𝐥𝐥 𝐜𝐮𝐥𝐭𝐢𝐯𝐚𝐭𝐢𝐨𝐧. We introduce sucrose for plant cell cultivations as a completely new substrate in our Feeding Technology portfolio. As a start, Feed Beads with 12 mm diameter for shake flask cultivations, Feed Plates with 96 round wells, and Feed Plates with 24 or 96 square wells are available with sucrose as slow-release fed-batch substrate. The products are available with different release rates of sucrose and have a linear release profile over the course of at least 96 hours. Adaptations of the release rates (to a certain extent) are possible, depending on the customer’s requirements.
