The buildup of misfolded protein aggregates is a key feature of neurodegenerative diseases like Alzheimer's. These aggregates contribute to a toxic cellular environment, driving disease progression. Identifying the specific proteins involved is essential for developing effective treatments. However, despite current knowledge, our understanding of the proteome within Aβ aggregates and their interactions remains incomplete. Existing spatial proteomics methods often rely on antibody arrays, limiting de novo proteomic discovery with precision. At Neuroscience 2024, our scientist CHIEN-CHANG HUANG will present findings from a study using the Microscoop system. This research uncovered both known and novel Aβ-associated proteins, including two newly identified proteins that colocalize with Aβ1-42. These findings were confirmed by antibody staining and further validated using brain sections from the 5XFAD mouse model of familial Alzheimer's disease. Can't make it to the event? No worries! You can still access the poster by reaching out to us. Struggling with proteomic challenges in neuroscience? You're not alone. Precise localization of proteins at the subcellular or synaptic level often proves difficult, limiting insights into neural processes. Detecting low-abundance proteins without interference from more abundant ones is also a significant hurdle. Brain tissue preservation methods, such as FFPE, can compromise data quality, making protein extraction harder. Identifying disease-specific biomarkers for conditions like Alzheimer's and Parkinson's is further complicated by overlapping protein profiles, while current spatial proteomics methods, relying on pre-defined antibodies, add both cost and time to the process. If these challenges resonate, Microscoop® by Syncell offers a solution. It enables de novo protein identification from specific cell compartments and microenvironments, directly extracting proteins for mass spectrometry analysis to reveal novel proteins with subcellular precision and uncover new insights into cellular processes. 📢 Join us for a product theater talk with Dr. Jung-Chi Liao at Neuroscience 2024 on October 7th, 2024, at 1 PM, and learn how Microscoop® can overcome these obstacles and accelerate your research. Looking forward to seeing you there!🙌 #SfN2024 #aggregates #biomarkerresearch #targetidentification #Syncell #Neuroscience2024

Misfolded protein aggregates are a hallmark of neurodegenerative conditions like Alzheimer's, Parkinson's, ALS, and Huntington's disease, creating toxic environments that drive disease progression. For example, Alzheimer's disease is marked by β-amyloid plaques and tau tangles that disrupt brain function, while Parkinson's disease features α-synuclein aggregates known as Lewy bodies. Understanding these protein aggregates is crucial for developing effective treatments for these challenging conditions. The blog explores a new spatial proteomics discovery technique that enables researchers to pinpoint the exact location and composition of protein aggregates within diseased tissues. Alzheimer's disease (AD) is a progressive neurodegenerative disorder that affects behavior, cognition, learning, and memory in older adults. Research led by Dr. Rodrigue at the Center for Vital Longevity shows that Aβ burden increases steadily from ages 30 to 89 in healthy individuals. Despite this knowledge, our understanding of the proteome of Aβ aggregates and their interactions with associated proteins remains incomplete. Current spatial proteomics methods often rely on antibody arrays, limiting de novo proteomic discovery with precision. At the FASEB conference on Cellular and Molecular Mechanisms of Brain Aging, our scientist Mei Zhang will present findings from a study utilizing the Microscoop system, which uncovered both known and novel Aβ-associated proteins. Among these, two newly identified proteins were found to colocalize with Aβ1-42, confirmed through antibody staining and further validated using brain sections from the 5XFAD mouse model for familial Alzheimer's disease.
