#Evizia has been posting on PRECYSE’s primary application of NGS QC. One of our early access collaborators asked if we could see the polymerase bound to the DNA before sequencing. Indeed, this is something PRECYSE can do very well. Per a 2017 Nature Communications paper https://lnkd.in/eP2FYiFF Evizia’s co-founder Dr. Jason Reed et al. published a DNA mapping technique using CRISPR-Cas9, whereby the protein is clearly visible due to its significantly larger z-axis height compared to DNA. This technique was also used as an early confirmation of linear DNA sizing with the instrument. We imaged several #Oxford_Nanopore_Technologies (#ONT) and #PacBio fully constructed libraries from collaborators with and without either the helicase (ONT) or polymerase (PacBio) addition before sequencing. All images generated by PRECYSE can be viewed in 2D or 3D, whereby the z-axis height of molecules from the consumable background surface is color coded. One of many really cool capabilities of PRECYSE! …and maybe this will unlock our ability to better optimize NGS enzyme concentration and binding for sequencing.

#Evizia continues to work with early access collaborators running Long Read Sequencing (LRS) library samples for #PacBio and #OxfordNanopore (#ONT) on the PRECYSE instrument. We’ve been gathering feedback on how best to display sizing information. With this innovative technology we are directly imaging and measuring 40-800+DNA molecules from a given sample, thus the data is not indirect (DNA ladder) RFU mass-based sizing everyone is used to seeing from electrophoresis systems. In the LRS world small molecule populations can be a problem and are severely underrepresented in electropherograms due to small mass (limited RFUs). Unanimous feedback, and we agree, is a sizing histogram that allows direct comparison to read length N50 and mean insert size data generated by LRS instruments. Here is a sample provided by the #BROAD_Institute where the Femto generated a mean of 43.7kbp. PRECYSE showed a 12.2 kbp mean correlating nicely with the ONT N50 of 14.6 kbp. We consistently see high percentages of 1-5kbp DNA in LRS libraries, even after many commonly used size ‘clean-up’ techniques.

Evizia has been working with several leading US Academic #genomics research cores to further test the PRECYSE instrument with gDNA and #NGS library samples. The image below is a #PacBio library provided by a collaborator in Boston. A really clean beautiful library! Sizing comparison to Agilent #Femto: PRECYSE: 431 molecules analyzed in a 4min run (150+ molecules typically needed for statistically accurate sizing) Mean size = 9.0kb Femto: max RFU peak = 13.9kb. Avg insert size from PacBio sequencing data = 10.2kb. We consistently see that sizing by electrophoresis utilizing fluorescence overestimates the actual size of DNA, increasingly so for larger molecules.
