Lactate dehydrogenase A (LDHA) promotes glioblastoma progression by facilitating interaction between tumor cells and macrophages. Inhibiting LDHA disrupts this interaction, suppressing tumor growth and reducing macrophage infiltration. The mechanisms underlying #enhanced #immunotherapy efficacy through #exercise. (A) The #antitumor effects of exercise depend on IL-15 signaling pathway, thus enhancing the sensitivity of pancreatic tumor cells to immunotherapy. (B) Exercise increases the infiltration of CD8+ T cells through the CXCL9 or CXCL11/CXCR3 signaling pathway. (C) Exercise influences multiple transcriptional pathways, including promotion of vascular branching and normal vasoconstriction, which improves hypoxia in TME and enhances the efficiency of drug transport.

In pancreatic ductal adenocarcinoma (PDAC), glutamine (Gln) is vital for cell proliferation and redox balance. Yet initial efforts to inhibit its metabolism did not succeed--they encountered issues of metabolic reprogramming and therapy resistance. Joel Encarnación-Rosado et al. identified 6-diazo-5-oxo-L-norleucine (DON) and its pro-drug version, sirpiglenastat (DRP-104), as effective Gln antagonists. Their research shows that DON induced a metabolic crisis in vitro, and DRP-104 significantly reduced tumor growth in vivo. NOW ONLINE This study identified EN1 and TGFβ-EN1 signaling as important features of #AdenoidCysticCarcinoma highlighting the diagnostic promise of EN1 🔓 OPEN ACCESS ➡ https://lnkd.in/eke6S7Av Accurate diagnosis of adenoid cystic carcinoma (ACC) often requires distinction from basal cell adenoma (BCA). Diagnostic markers capable of differentiating ACC from BCA, however, are lacking. Here, the authors investigated the diagnostic potential of homeobox transcription factor EN1. Immunohistochemistry revealed elevated EN1 expression specifically in ACC, enabling differentiation from BCA. Experiments using a translational patient-derived organoid model indicate that EN1 is regulated through TGFβ signaling, wherein TGFβ-induced EN1 promotes tumor budding, and that ACC develops invasive growth characteristics via tumor budding. The findings identify EN1 and TGFβ-EN1 signaling as important features of ACC and highlight the diagnostic promise of EN1.
