#DYK that #OnThisDay in 1956 Gregg Leonard Semenza was born, an American biochemist, geneticist and oncologist who shared the 2019 Nobel Prize in Physiology or Medicine with William G. Kaelin Jr. and Peter J. Ratcliffe, for their work on how cells sense and adapt to oxygen availability. Semenza made the ground-breaking discovery of the HIF-1 (hypoxia-inducible factor 1), a protein, which controls genes in response to changes in oxygen availability, and adjust how much oxygen is received by cells. This line of investigation has far-reaching implications in the treatment of various diseases, including cancer, heart attacks, anemia and strokes. For example, further study can be made to examine if cancer cells are seeking oxygen when they spread into surrounding tissues and the bloodstream which carries them around the body. https://lnkd.in/eKmQSGf7

Our latest paper in collaboration with Martin Ulmschneider is now available to read in ACS Omega. In this manuscript, Zhiwen Zhong has combined coarse-grain and all-atom molecular dynamics simulations to identify the key pairs of residues in the transmembrane domains of the TREM2 and DAP12 proteins that interact in order to stabilise the complex that the two proteins form. As mutants of TREM2 have been found to be risk factors for Alzheimer's Disease, we are hoping that these findings may be useful in identifying novel strategies to fight this (and potentially other) neurodegenerative disease(s). 👀 https://lnkd.in/emPYfB4M

SCIENCE SPOTLIGHT! 👨🔬🔬 Michelle Lance, Ph.D., Associate Computational Biologist III, under Dr. Nate Tucker, is working on the genetic contribution to cardiovascular disease. Her studies are offering insight into how changes at the cellular level can affect heart function or disfunction. Her latest project harnesses the power of natural variation in the human population in the form of single nucleotide polymorphism, or SNPs, to identify potential genetic contributors to cardiovascular disease. Excited to finally share my work on interrogating the functions of the BRCA pathway in somatic cell reprogramming! 🎉 Using separation-of-function mutations in SFP (stalled fork protection), RGS (replication gap suppression) and HDR (homology-directed repair), we show that iPS cell generation depends on the HDR-mediated repair of replication-associated DSBs. Enhancing HDR through ablation/down-regulation of 53BP1 further improves reprogramming in mouse and human cells. Dr. Hemali Phatnani, Ph.D., Assistant Professor of Neurological Sciences at Columbia University and Core Faculty Member and Director of the Center for Genomics of Neurodegenerative Disease (CGND) at the New York Genome Center, is working to answer elusive questions that can transform our understanding of ALS-FTD. 🧠 Dr. Phatnani’s research (funded by Target ALS) will decode which specific genes change in healthy or sick neurons and neighboring cells within ALS and FTD cases.
