Grant Award Alert! We are thrilled to announce that GenNext Technologies has received a $2.5 million dollar grant from NIGMS to fund the development of advanced protein footprinting technology and products that support multiple labeling chemistries with companion real-time radical dosimetry and custom data processing software. This marks our twelfth SBIR award with aggregate funding in excess of $12 million dollars! Collaborating with the laboratories of Professors Michael Gross (Washington University in St. Louis) and Mark Chance (Case Western Reserve), our “Multi-Radical Protein Footprinting Platform (MRPF)” will extend radical labeling to challenging protein regions, such as the transmembrane domain of membrane protein targets, including G-protein coupled receptors. The company plans to introduce MRPF technology as future upgrades to its exclusive, first-in-class SamFox® and AutoFox® radical protein footprinting products. We are grateful to the National Institute of General Medical Sciences for its continued support and acknowledgement of our team’s effort to advance and facilitate structural biology research. The Research Corporation for Science Advancement chose Professor Aaron Leconte as one of 11 researchers to receive a 2024 Cottrell Plus SEED Award! Leconte, an associate professor of chemistry, is studying chemically modified forms of DNA (known as "XNA”). Leconte’s project is titled “Towards XNA PCR: Evaluating the Potential of Fusion Domains to Improve XNA Polymerases.” Leconte is exploring how to better synthesize XNAs. Well-characterised, selective small molecules are essential for understanding the cellular functions of proteins and validating them as drug targets. Conversely, the use of poorly evaluated molecules or the biased interpretation of data generated with promiscuous substances leads to pollution of the literature with unsubstantiated claims. The ELRIG Drug Discovery in Germany forum will focus on data quality generated with small molecules and present best practices and guidelines for the use, availability and characterisation of these tools using state-of-the-art methods. We will highlight examples with both traditional inhibitors and novel chemical modalities, such as PROTACs. The meeting will bring together approaches from industry and academia, working together to improve the reproducibility and robustness of scientific data, thereby advancing target validation and our understanding of target biology. #ACHEMA24 #ELRIG

The throughput of ion channel recordings increased to unprecedented levels with the use of #AutomatedPatchClamp. To make full use of such high-throughput #electrophysiology technology, academic labs have often pooled resources in a shared, core #APC facility. One of the earliest adopters of a core QPatch facility was The University of Queensland shared by the labs of Irina Vetter, Thomas Durek, Fernanda Cardoso & Glenn King. They have now published almost 50 #QPatch papers in the past decade and have been keeping the printing-presses busy in 2023 with a flurry of papers characterizing peptide #Toxins modulating sodium #IonChannels and the discovery of #TMEM233 – a new Nav1.7 accessory protein. These studies will provide further tool molecules and novel protein targets to understand and develop therapeutics for #ChronicPain.   Congratulations to all authors. As we approach the end of 2023, I want to extend warm holiday greetings to everyone and express my deep gratitude to our GenNext® team and clients. It's a great time to look back at our remarkable scientific achievements this year. A highlight was at ASMS, where our seminal product publication, “Flash Oxidation (Fox®) System: A Novel Laser-Free Fast Photochemical Oxidation Protein Footprinting Platform,” received the prestigious ASMS Ron Hites Award,  underscoring the significant impact our Fox Technology is making in the scientific community. Excitingly, we also unveiled our latest innovation, the AutoFox® System (https://lnkd.in/gVaUTE-4). This first-in-class product is set to revolutionize structural biology research by automatically labeling samples in a 96-well format, enhancing efficiency and precision. Additionally, our Scientific Advisory Board made significant contributions with numerous publications. Their work has been instrumental in establishing radical protein footprinting as a premier technique in structural biology. Notable papers include Michael Gross’s “Characterization of Higher Order Structural Changes of a Thermally Stressed Monoclonal Antibody via Mass Spectrometry Footprinting and Other Biophysical Approaches,” a collaborative effort from Joshua Sharp and Lisa Jones entitled “Dimethylthiourea as a Quencher in Hydroxyl Radical Protein Footprinting Experiments,” and Mark Chance’s insightful “Intact Mass Spectrometry Screening to Optimize Hydroxyl Radical Dose for Protein Footprinting.” Once again, I extend my sincere thanks to all of you for your unwavering commitment and outstanding contributions to the field of structural biology.
