My talk will focus on ultra-low volume #liquidhandling for screening applications and how these techniques can significantly improve precision and efficiency in the laboratory.     Precise dispensing at the picoliter and nanoliter scale is not just a technique - it is a transformative tool that can revolutionize the way we approach scientific work. Why am I so passionate in creating those videos? Because I want to show how precise and fast microdispensing can help innovation in: 💧 Assays for diagnostics 💧 Dispense on Microfluidics 💧 Multiplex immunoassays 💧 Lateral flow applications 💧 Coating biosensors/biochips 💧 Protein and DNA microarrays 💧 Reverse-phase protein arrays (RPPA) And so much more…   In addition to improving experimental accuracy and reproducibility, these techniques are opening up new opportunities to explore complex biological processes and develop targeted therapeutics.   Developing our microarray spotters at M2-Automation GmbH and M2-Instruments GmbH is more than just a job to me, and I believe these images reflect my deep-rooted passion and emotional connection to my work – my life. I hope that my insight into this fascinating technology will inspire you to explore the endless possibilities it offers.   Details: All videos are real-time footage showing actual pico- and nanoliter dispensing operations without rendering, using AI or other manipulation. For the video enthusiasts among you, for the parts of the video where the actual dispensing nozzles are shown, I used a Sony alpha 7iii with a macro lens. To examine the microarrays inside the 96-well plates, I used my favourite toy, the 3D microscope from Microqubic.   By the way: My favourite is the one where I'm trying to pipette by hand next to the dispenser in a very shaky way.     For microarray production, there are cases where non-contact spotting is not the optimal solution. When dealing with heterogeneous and viscous samples like cell lysates, contact pin spotting truly excels. Additionally, when thousands of source samples need to be spotted, a large multi-print head is invaluable.   These two requirements come into play when Biobank samples need to be protein-profiled, using for example a reverse-phase protein array (RPPA) investigation approach. At M2-Automation GmbH, we've significantly enhanced our pin spotting options, hardware, and software over the past few years. We now offer various pin head formats that can also accommodate your existing pin inventory.   👏 A special shoutout to INTER-ARRAY by fzmb GmbH, a close partner of M2-Automation GmbH, for their major influence on the further development of our pin spotting technology. The video was created by INTER-ARRAY by fzmb GmbH, showcasing the advancements and collaborative efforts that have driven our innovations forward. 👌   Details of the video: It shows our standard 36-pin head fitted with only 12 blunt-end pins. The pins have to be re-inked after each dispensing and washed after each sample change. The microdispensing instrument is an iONE-1200 which holds 140 slide sized targets on a vacuum table.     A perfect combination: high-speed dispensing meets glass slides with functionalized surfaces that capture liquids without physical barriers!   Hydrophilic areas enclosed by a superhydrophobic surface allow precise control over the size, shape and placement of the dispensed microdroplets. On contact, the liquid is naturally drawn into the hydrophilic area. Droplet migration is prevented by local differences in wettability. The geometry of the spot, whether circular or rectangular, is determined by the surface design and not by the interaction between the liquid and the wetted surface. ⚡ In short, these hydrophilic anchors make life incredibly easy! ⚡   Why should you choose DMA-Slides from Aquarray? 💧 Droplet volume per spot from 10 nl to 5 µl 💧 Reproducible arrays/grids on slides streamline automated readouts 💧 Dispensed droplets are held stably in place due to the superhydrophobic background, improving reproducibility during sampling 💧 Wide range of patterns resulting in a range of 80 to 6048 spots per DMA, exceeding the throughput of microplates 💧 Standard microscope slide format with options for different coatings and spot geometries   Application example: biological assays ✔  Cell-based screenings ✔ Microscopic readout, e.g. fluorescence, morphology ✔ The number of cells required is low, between 1 and 5000 cells, depending on spot size ✔ Many experiments with rare cell types possible (biopsy) ✔ On-chip compound libraries pre-printed on the array   Details of the video: A DMA-Slide “203” from Aquarray was printed with a nanoliter dispenser on an iONE-1200 from M2-Automation GmbH. One shot with water, one with coloured ink, about 150 nanoliters per spot. Like all my videos… not rendered or AI generated and of course at real speed.     Why is nanoliter-dispensing in a multi-channel setup so sought after for production systems in diagnostics and life sciences applications? It`s versatility across surfaces, sample arrangement, pattern geometry, droplet volume and amount of droplets helps:   ✔ Saving time, investments and valuable lab space on our customers site. ✔ Miniaturization… hundrets of experiments on just one microfluidic cartridge or on a small piece of a membrane ✔ Enabling the production of very small biochips/biosensors ✔ Small droplets enable reliable imaging/counting because they are highly reproducible objects dominated by surface tension ✔ Instead of adding 1 uL at one position, adding 10 x 100 nL or 100 x 10 nL often delivers better results and allows more flexibility ✔ Saving time and money by making tests smaller and using less expensive materials   Details of the video…   🚿 First part (dispensing): Droplet of 15 nl each dispensed with an iONE-1200 from M2-Automation GmbH in a 3-channel setup in stop-to-spot mode (the dispensing nozzle stops for each droplet ejection) on a “Rhombic chamber chip eP1” from microfluidic ChipShop GmbH. The last droplet was produced by dispensing 30 x 15 nl with a frequency of 20 droplets/second forming that beautiful piece of art. The distance between the droplets is about 700 µm, one chamber of the chip is 6500 µm wide resulting in a diameter of 5500 µm of the biggest red circle. Green ink was aspirated into channel 1, red ink into channel 2 and 3 showcasing the flexibility (3 different samples or 3 identical samples per aspiration cycle).   Fun fact: we had ChatGPT creating a CSV-file with the X and Y coordinates to dispense on according to our specifications. Those files were then imported into the control software InDot (via the software feature “CSV Import”) and applied to the target… creating arbitrary pattern geometries on the target surface within a few minutes and lots of fun! 😁 I love my job!   🔬 Second part (inspection): After letting the ink dry a little the chip was placed into the 3D Microscope from Microqubic for a closer inspection of the chip and the droplets dispensed. What a great tool to make the hidden details visible. Give the run a name, save it, and start immediately. The video shows an example of a typical run designed in InDot. ✅ It starts with a 𝘞𝘢𝘴𝘩𝘪𝘯𝘨 tile.  ✅ That’s followed by a tile for an individual 𝐴𝑠𝑠𝑖𝑔𝑛𝑒𝑑 𝑃𝑎𝑡𝑡𝑒𝑟𝑛.  ✅ Then comes an imaging step of your spotted features, called 𝘗𝘪𝘤𝘵𝘶𝘳𝘦𝘴 𝘰𝘧 𝘍𝘦𝘢𝘵𝘶𝘳𝘦𝘴. ✅ Then we have the penultimate stage of 𝑀𝑜𝑣𝑖𝑛𝑔 𝑡𝑜 𝑃𝑜𝑠𝑖𝑡𝑖𝑜𝑛, which in this case is 𝘏𝘰𝘮𝘦. ✅ Final stage: Name, save and start the run.    It couldn’t be simpler.   𝘐𝘮𝘱𝘰𝘳𝘵𝘢𝘯𝘵 𝘵𝘰 𝘯𝘰𝘵𝘦: Before designing a run, all necessary tasks, e.g. sample handling, source selection, wash-step design, target and pattern design must be completed in advance and then placed in the correct chronological order, in the form of tiles.
