Alzheimer's disease (AD) is a progressive neurodegenerative disorder that affects behavior, cognition, learning, and memory in older adults. Research led by Dr. Rodrigue at the Center for Vital Longevity shows that Aβ burden increases steadily from ages 30 to 89 in healthy individuals. Despite this knowledge, our understanding of the proteome of Aβ aggregates and their interactions with associated proteins remains incomplete. Current spatial proteomics methods often rely on antibody arrays, limiting de novo proteomic discovery with precision. At the FASEB conference on Cellular and Molecular Mechanisms of Brain Aging, our scientist Mei Zhang will present findings from a study utilizing the Microscoop system, which uncovered both known and novel Aβ-associated proteins. Among these, two newly identified proteins were found to colocalize with Aβ1-42, confirmed through antibody staining and further validated using brain sections from the 5XFAD mouse model for familial Alzheimer's disease. Misfolded protein aggregates are a hallmark of neurodegenerative conditions like Alzheimer's, Parkinson's, ALS, and Huntington's disease, creating toxic environments that drive disease progression. For example, Alzheimer's disease is marked by β-amyloid plaques and tau tangles that disrupt brain function, while Parkinson's disease features α-synuclein aggregates known as Lewy bodies. Understanding these protein aggregates is crucial for developing effective treatments for these challenging conditions.
