Tips and Tricks for Cell-Based Imaging Assays: Immunocytochemistry Overview Immunocytochemistry (ICC) is a powerful method employed to visualize specific proteins or antigens within cells. It harnesses the binding specificity of antibodies in conjunction with immunofluorescence microscopy, enabling the examination of subcellular structures. The ICC process involves a series of critical steps, namely fixation, permeabilization, blocking, antibody binding, and washing, all of which are essential for attaining accurate and reliable results. The execution of ICC in 96-well and 384-well plates is sometimes informally referred to as an "In-Cell Western," although "In-Cell Western™" is a trademark of LI-COR Biosciences. A recent study on enhancing antibody production by hybridoma cells using functionalized parylene films was published in ACS Applied Bio Materials journal. We express our deepest gratitude to the collaborative teams led by Prof. Jae-Chul Pyun and Prof. Misu Lee for their outstanding joint achievements. The team found that the chemical functional groups of nitrogen and oxygen species influenced the enhanced attachment of hybridoma cells to the surface of the culture plate, leading to improved antibody production. Additionally, they identified genes associated with antibody production that were up-regulated in hybridoma cells cultured on modified parylene films. In this study, the hybridoma cells on various parylene films were monitored and captured every 12 hours for 72 hours using our automated live-cell imaging system, Celloger® Mini. 📚 PUBLICATION | Blood-brain barrier #organoids grown in our #Gri3D 600 µm microwells Employing receptor-mediated transcytosis (RMT) emerges as an effective approach for delivering biologic agents across the blood-brain barrier (#BBB). The recent progression in creating human BBB organoid models is a significant breakthrough, aiding in understanding RMT mechanisms and expediting the development of brain delivery technologies for #drugdevelopment. However, the existing techniques for generating organoids suffer from multiple limitations, preventing the use of #BBBorganoids as a screening tool 💡 Elena Kassianidou, Ph.D. and colleagues from Roche describe in their protocol the use of #Gri3D hydrogel-based arrays for #BBBorganoids.
