    Microscoop, an innovative microscopy platform, is designed to fill the needs of identifying novel protein constituents at the protein-protein interaction level. Existing mass spectrometry-based unbiased methods often lack subcellular spatial precision or sensitivity for high-precision proteomic discovery, both for cell and tissue samples.  Our solution is "optoproteomics," a cutting-edge technique that combines microscopy, mechatronics, photochemistry, and advanced image processing (either deep learning or traditional methods) to enable ultrahigh-content in situ photo-biotinylation. The Microscoop system photolabels proteins at user-defined regions of interest (ROIs) under a microscope, utilizing directed photochemistry one field of view (FOV) at a time for tens of thousands of FOVs with similar morphological features. Here’s how it works: 1) Microscopy Integration: The system uses advanced microscopy to visualize and define ROIs within cell or tissue samples. 2) Directed Photochemistry: Utilizing precise optics and photochemistry, the system photolabels proteins within these ROIs at 240-nm resolution. 3) High-Throughput Capability: This process is repeated across tens of thousands of FOVs, overcoming protein amplification problem by protein accumulation. 4) Extraction and Analysis: Photolabeled proteins are then extracted and analyzed via mass spectrometry, revealing both known and novel protein players specific to the ROIs.
