🧠 Groundbreaking research offers new insights into Alzheimer’s disease (AD) progression. A study suggests that glucose metabolism in the posterior cingulate cortex (PCC), measured by the standard uptake value ratio (SUVR) from FDG PET imaging, could serve as a supplementary biomarker alongside amyloid-β (Aβ) and tau. Kaplan-Meier survival analyses indicate that combining PCC SUVR with CSF Aβ42 and ptau-181 enhances the predictive performance for the progression from cognitively unimpaired (CU) to AD. Huntingtin contains an ubiquitin-binding domain and regulates lysosomal targeting of mitochondrial and RNA-binding proteins The authors demonstrate that HTT physically interacts with ubiquitin and co-purifies with several proteins involved in RNA metabolism, stress granule formation, and protein quality control, suggesting a broader function than previously ascribed. Notably, HTT's interaction with these proteins is enhanced by polyglutamine expansion and attenuated by phosphorylation at serine 421, implying a potential mechanism for disease-associated dysfunction. Functional studies reveal a critical role for HTT in mitochondrial autophagy, as evidenced by reduced lysosomal mitochondrial content in HTT knockout cells. Additionally, the accumulation of RNA-binding proteins in lysosomes of HTT-deficient cells suggests a compensatory upregulation of macroautophagy in response to cellular stress. These findings position HTT as a key regulator of proteostasis with implications for Huntington's disease pathogenesis. A nutrient-dependent molecular feedback circuit converts lysosomes into organelles that support anabolism or catabolism. Researchers have discovered a mutually exclusive molecular switch between PI(4)P and PI(3)P, which rapidly recalibrates lysosome function. This recalibration optimally aligns with the catabolic or anabolic functions that best support the cell's nutrient state, as influenced by mTORC1 activity. This adaptive process takes place before slower, longer-term responses related to biogenesis. This process might have implications for the therapeutics of diseases with lysosomal dysfunction, such as Lysosomal Storage Disorders, Alzheimer's and Parkinson's.
