Huntingtin contains an ubiquitin-binding domain and regulates lysosomal targeting of mitochondrial and RNA-binding proteins The authors demonstrate that HTT physically interacts with ubiquitin and co-purifies with several proteins involved in RNA metabolism, stress granule formation, and protein quality control, suggesting a broader function than previously ascribed. Notably, HTT's interaction with these proteins is enhanced by polyglutamine expansion and attenuated by phosphorylation at serine 421, implying a potential mechanism for disease-associated dysfunction. Functional studies reveal a critical role for HTT in mitochondrial autophagy, as evidenced by reduced lysosomal mitochondrial content in HTT knockout cells. Additionally, the accumulation of RNA-binding proteins in lysosomes of HTT-deficient cells suggests a compensatory upregulation of macroautophagy in response to cellular stress. These findings position HTT as a key regulator of proteostasis with implications for Huntington's disease pathogenesis. 🧠 Groundbreaking research offers new insights into Alzheimer’s disease (AD) progression. A study suggests that glucose metabolism in the posterior cingulate cortex (PCC), measured by the standard uptake value ratio (SUVR) from FDG PET imaging, could serve as a supplementary biomarker alongside amyloid-β (Aβ) and tau. Kaplan-Meier survival analyses indicate that combining PCC SUVR with CSF Aβ42 and ptau-181 enhances the predictive performance for the progression from cognitively unimpaired (CU) to AD. New findings from a study comparing normal brain samples to those from patients with ALS reveal important insights into the disease. The study utilized the new HTRF Total Ataxin 2 and the Mutant Ataxin 2 Detection Kits to analyze protein samples. Results showed that the Ataxin 2 protein was more highly expressed in the ALS patient sample, with a high aggregation rate. Notably, the mutant form of the protein was only detectable in the ALS patient sample, and a disaggregation step resulted in a positive assay signal. These findings provide a better understanding of the role of Ataxin 2 in ALS and could lead to new approaches for treating the disease.
